Stem cell niche is critical for regulating the behavior of stem cells. Drosophila neural cells (Neuroblasts, NBs) are encased by glial closely, but it still remains unclear whether can regulate self-renewal and differentiation NBs. Here, we show that ferritin produced glia, cooperates with Zip13 to transport iron into NBs energy production, which essential proliferation The knockdown encoding genes causes shortage in via downregulating aconitase activity NAD + level, leads low premature mediated Prospero entering nuclei. More importantly, a potential target tumor suppression. In addition, level production affected status NBs, establishing bicellular homeostasis. this study, demonstrate indispensable maintain unveiling novel role NB during brain development.

ABSTRACT Extracellular vesicles (EVs) are secreted nanoparticles composed of a lipid bilayer that carry lipid, protein, and nucleic acid cargo between cells as mode intercellular communication. Although EVs can promote tissue repair in mammals, their roles animals with greater regenerative capacity not well understood. Planarian flatworms capable whole‐body regeneration due to pluripotent somatic stem called neoblasts proliferate response injury. Here, using transmission electron microscopy, nanoparticle tracking analysis, protein content examination, we showed enriched from the tissues planarian Schmidtea mediterranea had similar morphology size other eukaryotic EVs, these carried orthologs conserved EV biogenesis regulators ALIX TSG101. PKH67‐labeled were taken up efficiently by cells, including S/G2 neoblasts, G1 neoblasts/early progeny, differentiated cells. When injected into living planarians, regenerating fragments enhanced upregulation neoblast‐enriched proliferation‐related transcripts. In addition, injection increased number F‐ara ‐EdU‐labelled 49% compared buffer only. Our findings demonstrate planarians produce cell proliferation, suggest an amenable vivo model for study function during regeneration.
