Abstract The evolution of complete reproductive isolation hinges on the cumulative action isolating barriers that can manifest throughout life cycle an organism. Consequently, a comprehensive understanding features underlying origin and maintenance species requires assessing relative contributions distinct to overall isolation. Here we characterize multiple interrelated across various developmental stages nematode sister Caenorhabditis remanei latens . We quantified F1 hybrid male sterility characterized phenotypic causes associated with abnormalities in germline as well non-germline gonad somatic tissues, uncovering complex suite defects contributing strong postzygotic Despite these multifarious barrier traits, assays testing for interspecies sperm transfer under “choice” conditions did not yield evidence premating In contrast other pairs, also found no ectopic migration acts postmating-prezygotic barrier. constellation hybrids points polygenic or highly pleiotropic basis dysfunction implicates more rapid intrinsic than prezygotic organisms.

Given the interest in biogeography and diversity of Caenorhabditis genus, we established a collection these nematodes from field surveys on four Indonesian islands. We isolated over 60 strains belonging to ten species. Five species were previously known other locations: C. briggsae , which was predominant, tropicalis, nigoni, brenneri elegans . The five are new, describe them here as indonesiana, malino, ceno, brawijaya ubi RNA sequence analysis 1,861 orthologous genes placed all Indonesia Elegans group Four new belong Sinica subclade so far only found an East Asia-Indo-Pacific world region. fifth species, indonesiana appears sister tropicalis - wallacei pair, both also Indonesia. present findings thus consistent with diversification having occurred this Crosses between closely related showed counterexamples Haldane's "rule": for two pairs one cross direction hybrid males. In addition, pair that could partially interbreed: (East Java) C sp. 41 (Solomon islands), males being fertile. Such closesly good models genetic studies incompatibilities arising during speciation. This work addresses their biodiversity, phylogenetic relationships nematodes, laboratory model organisms. Through studies, authors Indonesia, representing including new. From sequencing reconstruction, crosses progeny can be females, abiding by rule, but cases fertile hybrids produced direction. These studying incompatibilities.
