The silky shark, Carcharhinus falciformis , is a cosmopolitan species commonly caught as bycatch for longline fisheries. However, the genetic stock structure Indo-Pacific Ocean not well-defined yet. Here, we used eight microsatellite loci to examine and effective population size of 307 sharks across 5 sampling locations. Considerable research exists on the life history traits, evolutionary history, and environmental factors that shape population genetic structure of marine organisms, including sharks rays. Large-bodied pelagic sharks are key regulators of oceanic ecosystem stability, but highly impacted by severe overfishing. One such species, the shortfin mako shark (Isurus oxyrinchus), a globally widespread, migratory predator, has undergone dramatic population reductions and is now Endangered (IUCN Red List), with Atlantic Ocean in particular assessed fishery managers as overfished need urgent, improved management attention. Genomic-scale assessments for this apex predator species have not been previously available to inform planning; thus, we investigated genetics across using bi-organelle genomics approach. Complete mitochondrial genome (mitogenome) sequences genome-wide SNPs from distributed revealed contrasting patterns structure marker types. Abstract Understanding the population dynamics of highly mobile, widely distributed, oceanic sharks, many which are overexploited, is necessary to aid their conservation management. We investigated global genomics tiger sharks (Galeocerdo cuvier), a circumglobally apex predator displaying remarkable behavioral versatility in its diet, habitat use (near coastal, coral reef, pelagic), and individual movement patterns (spatially resident long-distance migrations). genotyped 242 from 10 globally distributed locations at more than 2000 single nucleotide polymorphisms. Although this species often conducts massive distance migrations, data show strong genetic differentiation both neutral (FST = 0.125–0.144) candidate outlier loci 0.570–0.761) between western Atlantic Indo-Pacific suggesting potential for adaptation environments specific these regions. Within regions, there was mixed support northern southern hemispheres Atlantic, none structure within Indian Ocean. Notably, results demonstrate low level remote Hawaiian archipelago compared with Ocean 0.003–0.005, P < 0.01). Are we there yet? Benchmarking low-coverage nanopore long-read sequencing for the assembling of mitochondrial genomes using the vulnerable silky shark Carcharhinus falciformis DOI Creative Commons

Whole mitochondrial genomes are quickly becoming markers of choice for the exploration within-species genealogical and among-species phylogenetic relationships. Most often, 'primer walking' or 'long PCR' strategies plus Sanger sequencing low-pass whole genome using Illumina short reads used assembling chromosomes. In this study, we first confirmed that can be sequenced from long nanopore data exclusively. Next, examined accuracy long-reads assembled chromosomes when comparing them to a 'gold' standard reference chromosome short-reads sequencing.Using specialized bioinformatics tool, produced assembly silky shark C. falciformis with an average base coverage 9.8x.
