Chinese hickory (Carya cathayensis) is an important tree species for agriculture, but dry rot disease, caused by Botryosphaeria dothidea, threatens its viability. To study the interactions between and pathogen, transcriptomic sequencing was conducted on infected healthy tissues from field-grown hickory. Differential gene expression analysis identified key defense pathways genes activated during infection. The also explored roles of non-coding RNAs, such as lncRNAs circRNAs, in tree’s defense. results showed that early mid stages infection, defends itself through mechanisms like enhanced lignin synthesis increased peroxidase activity. Non-coding RNAs contribute to disease resistance reinforcing cell wall, increasing oxidase activity, promoting antibiotic-related secondary metabolites. Additionally, patterns at these differ significantly those late stage when most are suppressed, PR1 WRKY2 show a decline. These findings offer valuable insights into pathogenesis potential strategies improving resistance.
