Hydrogen peroxide (H2O2) overexpressed in mitochondria has been regarded as a key biomarker the pathological processes of various diseases. However, there is currently lack suitable mitochondria-targetable near-infrared (NIR) probes for visualization H2O2 multiple diseases, such PM2.5 exposure-induced lung injury, hepatic ischemia-reperfusion injury (HIRI), nonalcoholic fatty liver (NAFL), fibrosis (HF), and malignant tumor tissues containing clinical cancer patient samples. Herein, we conceived novel NIR fluorescent probe (HCy-H2O2) by introducing pentafluorobenzenesulfonyl sensing unit into hemicyanine platform. HCy-H2O2 exhibits good sensitivity selectivity toward H2O2, accompanied remarkable "turn-on" fluorescence signal at 720 nm. Meanwhile, stable ability permits monitoring up-generated level during mitophagy. Furthermore, using HCy-H2O2, have successfully observed an overproduced mitochondrial ambient HIRI, NAFL, HF models through imaging. Significantly, achieved both tumor-bear mice well surgical specimens patients, making promising tool diagnosis imaging-guided surgery.

Dyes with extended conjugate structures are the focus of extensive design and synthesis efforts, aiming to confer unique improved optical electronic properties. Such advancements render these dyes applicable across a wide spectrum uses, ranging from second-window near-infrared (NIR-II) bioimaging organic photovoltaics. Nevertheless, inherent benefits long conjugation often accompanied by persistent challenges like aggregation, fluorescence quenching, absorption blueshift, low stability poor water solubility. Herein, structural strategy termed "homo-dyad outer hydration layer" is introduced address problems, tailored for development imaging probes exhibiting absorption/emission wavelengths. This approach involves bringing two heptamethine cyanines together through flexible linker, forming homo-dyad structure, while strategically attaching four polyethylene glycol (PEG9) chains terminal heterocycles. imparts excellent solubility, biocompatibility, enhanced chemical, photo-, spectral dyes. Utilizing this strategy, biomarker-activatable probe (HD-FL-4PEG9-N) NIR-II fluorescent 3D multispectral optoacoustic tomography developed, its effectiveness in disease visualization. It can not only serve as an injectable acute kidney injury due high but also sprayable bacterial-infected wounds.

Photosensitizer-based phototherapies, including photodynamic therapy (PDT) and photothermal (PTT), offer safe treatment modalities for tumor ablation with spatiotemporal precision. After photons are absorbed, PDT creates localized chemical damage by generating reactive oxygen species (ROS), while PTT induces thermal damage. However, still faces hypoxic challenges, encounters issues related to heat resistance potential overheating. The combination of shows great as an effective anticancer strategy. By targeting lysosomes carefully designed phototherapeutic reagents combined phototherapy, rapid dysfunction cell death in cancer cells can be induced, showing promise treatment. Herein, two α–α-linked bisBODIPYs tetraphenylethene (TPE) moieties synthesized. These TPE-substituted expand the absorption into NIR range (λmaxabs/λmaxem ∼ 740/810 nm) confer aggregation-induced emission (AIE) activity (λmaxem 912 nm). Moreover, these self-assemble surfactant F-127 nanoparticles (NPs), which efficiently generate ROS (1O2 •OH) both solution cellular environments demonstrate superior conversion efficiencies (η 68.3%) along exceptional stability. More importantly, NPs showed lysosomal remarkable murine models, indicating their precision therapy.

Real-time visualization of endogenous enzymes not only helps reveal the underlying biological principles but also provides pathological information for cancer/disease diagnosis and even treatment guidance. To this end, enzyme-activatable fluorescence probes are frequently fabricated that turn their signals "on" exclusively at enzyme-rich region, thus enabling noninvasive real-time imaging interest molecular level with superior sensitivity selectivity. However, in a complex context, commonly used single (i.e., single-locked) may suffer from "false positive" healthy tissues be insufficient to accurately indicate occurrence certain diseases. Real-time monitoring of the lysosomal microenvironment using small-molecule probes is critical for understanding complex interactions between organelles and diseases associated with dysfunction. Most traditional fluorescent-dye-based lysosome rely on protonation nitrogen atom in morpholine unit to visualize by inhibiting photoinduced electron transfer (PET) effect. However, these often face selectivity issues within cellular microenvironment. For instance, classic hemicyanine dyes (HD) show nonspecific fluorescence responses liver due inappropriate pKa value, leading low imaging contrast risk false positives. Herein, a series novel pH/viscosity-activatable, lysosomal-targeted near-infrared (NIR) fluorescent were developed incorporating naphthalimide dye dyes. Logic-gated strategies represent a promising approach to achieving highly selective cancer therapies. In this work, we present LG-AB (Logic-Gated Aza-BODIPY), an OFF-ON photothermal therapy (PTT) agent designed selectively target cells. undergoes red-shift in its maximum absorbance wavelength when activated the tumor microenvironment, enabling molecule precisely generate heat cancerous tissue upon light irradiation. Unlike conventional activatable agents that rely on single biomarker, employs AND logic-gated design, where glucose transporter 1 (GLUT1) overexpression facilitates targeted cellular uptake cells, followed by activation through elevated glutathione (GSH) levels.
