Natural small-molecule drugs have been used for thousands of years the prevention and treatment human diseases. Most natural products available on market modified into various polymer materials improving solubility, stability, targeted delivery drugs. However, these nanomedicines formed based carriers would produce severe problems such as systemic toxicity kidney metabolic stress. In contrast, carrier-free by their self-assembly in water inherent advantages low toxicity, good biocompatibility, biodegradability. This review summarizes assembly process application products, which are mainly driven multiple non-covalent interactions, includes single-molecule assembly, bimolecular drug-modified organogels. Meanwhile, molecular mechanism involved different processes is also discussed. Self-assembly simulation structural modification or traditional Chinese medicine molecules using dynamics computer-assisted methods proposed, will lead to discovery more nanomedicine drug systems. Overall, this provides an important understanding strategy study multi-molecule nanomedicines.

Abstract The emergence of nanotechnology has significantly promoted the development medical field, and nanomedicines broadened application potential existing drugs. However, at present, many new types suffer from poor stability, uncertain safety, complex preparation processes, high costs. nano‐ Chinese herbal medicines (CHMs) provided ideas for nanomedicines. Nano‐CHMs are simple to prepare, easily accessible, possess stable reliable safety. This review highlighted nanoparticles with pharmacological activities important material basis CHM formula decoctions. Given composition formulas, article further presented nanoaggregates exosome‐like vesicle nanostructures in single herbs, self‐assembly effect natural compounds. For first time, this systematically introduced characteristics formulas herbs then compounds, nano‐aggregates unclear components structures self‐assembled supramolecular materials. It also summarized problems be solved research CHMs applications, aiming provide guidance modernization ‐related nanoparticles.

In recent years, significant advancements in nanotechnology have yielded remarkable improvements biomedical applications. Nanocarriers, harnessed from the principles of nanotechnology, garnered widespread utilization medicine delivery and diagnostics. However, progression nanocarriers has been hindered by two key challenges: low drug loading capacity potential for carrier-induced toxicity. To surmount these obstacles, rapid development expansion carrier-free systems (CFDDSs) composed pure drugs prodrugs emerged as a promising solution. Extensive endeavors undertaken to explore novel excipients, therapeutic agents, self-assembly processes, mechanisms, aimed at expanding horizons CFDDSs enhancing their efficacy. This comprehensive review provides an overview CFDDSs, elucidating mechanisms. Additionally, we examine diverse applications while shedding light on challenges ahead future clinical implementation CFDDSs. serves enhance our understanding intricate mechanisms governing nanoassembly formation fosters advancement expansive realm research.

Abstract Background Ulcerative colitis (UC) is a chronic intestinal inflammation, resulting in global healthcare challenge with no real specific medicine. Natural medicines are recognized as potential clinical alternative therapy, but their applications limited by poor solubility and low bioavailability. Results In this work, inspired the natural of ancient China, novel functional carbon dots derived from Magnetite Medicated Leaven (MML-CDs) were synthesized hydrothermal method, confirmed ultrasmall nano-size (3.2 ± 0.6 nm) Fe doped surface structure, thereby excellent gastrointestinal stability, remarkable capabilities eliminating ROS, highly biocompatibility. With external stimuli, oral administration MML-CDs demonstrated obvious alleviation to UC. Further experiments pointed that could improve hemostasis capability, suppress inflammation reactions oxidative stress, up-regulate expression tight junction proteins. Furthermore, also showed well regulation dysbiosis flora. Conclusion Overall, above evidence reveals green-synthesized can significantly alleviate bleeding, inhibit colon repair colonic barrier damage, further regulating flora microenvironment. Our findings provide an efficient therapy strategy for ulcerative colitis. Graphical
