Abstract Macrophages are immune cells belonging to the mononuclear phagocyte system. They play crucial roles in defense, surveillance, and homeostasis. This review systematically discusses types of hematopoietic progenitors that give rise macrophages, including primitive progenitors, erythro-myeloid stem cells. These have distinct genetic backgrounds developmental processes. Accordingly, macrophages exhibit complex diverse functions body, phagocytosis clearance cellular debris, antigen presentation, response, regulation inflammation cytokine production, tissue remodeling repair, multi-level regulatory signaling pathways/crosstalk involved homeostasis physiology. Besides, tumor-associated a key component TME, exhibiting both anti-tumor pro-tumor properties. Furthermore, functional status is closely linked development various diseases, cancer, autoimmune disorders, cardiovascular disease, neurodegenerative metabolic conditions, trauma. Abstract Pathological microenvironment of diabetes including hyperglycemia, excessive reactive oxygen species (ROS), and accumulation advanced glycation end products leads to high risk infection persistent inflammatory reaction, retarding biointegration implants. To address this issue, a dual‐catalysis system consisting Na 2 TiO 3 nanotubes with CeO nanodots polydopamine (PDA) cover is constructed on Ti implant manipulate ROS generation scavenging for antibiosis tissue regeneration. form heterojunction, in which vacancies (VOs) contribute the separation electron‐hole pairs under near‐infrared light (NIR) irradiation. In NIR mode, photocatalysis‐induced photothermal‐induced hyperthermia by ‐CeO heterojunction PDA synergistically kill bacteria efficiently. non‐NIR show excellent superoxide dismutase (SOD) catalases (CAT) like enzyme activities, they catalyze (e.g., ·O − H O ) thoroughly into benign O, relieving oxidative stress accelerating M2 macrophage phenotype. It helps biofunctional recovery endothelial cells fibroblasts, promoting regeneration around implants diabetic models. This work proposes promising approach manipulating treat regulate thereby improving performance microenvironment.

Diabetic foot ulcers (DFU) are chronic, refractory wounds caused by diabetic neuropathy, vascular disease, and bacterial infection, have become one of the most serious persistent complications diabetes mellitus because their high incidence difficulty in healing. Its malignancy results from a complex microenvironment that includes series unfriendly physiological states secondary to hyperglycemia, such as recurrent infections, excessive oxidative stress, inflammation, ischemia hypoxia. However, current common clinical treatments, antibiotic therapy, insulin surgical debridement, conventional wound dressings all drawbacks, suboptimal outcomes exacerbate financial physical burdens patients. Therefore, development new, effective affordable treatments for DFU represents top priority improve quality life In recent years, nanozymes-based therapy systems been attracting extensive interest integrating unique advantages nanomaterials natural enzymes. Compared with enzymes, nanozymes possess more stable catalytic activity, lower production cost greater maneuverability. Remarkably, many multienzyme activities can cascade multiple enzyme-catalyzed reactions simultaneously throughout recovery process DFU. Additionally, favorable photothermal-acoustic properties be exploited further enhancement therapeutic effects. this review we first describe characteristic pathological DFU, then discuss mechanisms applications healing, finally, highlight challenges perspectives nanozyme treatment.
