Abstract The cGAS‐STING signaling pathway is a pivotal immune response mechanism that bridges tumor and cell interactions. This study describes multifunctional LPDAM nanoplatform integrating Lapachone, polydopamine (PDA), Mn 2+ , which synergistically kills cells activates the pathway, thereby inducing DC maturation T activation to achieve potent antitumor immunity. In situ personalized tumor vaccines are produced directly at the primary site by killing cancer cells and stimulating immune cells, they effective against individuals bypass complexity high cost of in vitro vaccine production. However, their clinical application is hindered insufficient efficiency inducing immunogenic cell death (ICD) systemic inflammation caused adjuvants. Here, constructed for melanoma immunotherapy based on bioorthogonal catalytic microneedles, which enable release prodrugs sites mediate strong ICD an enhanced response while avoiding storms toxic side effects. By incorporating TiO2 nanosheets supported Pd into swellable microneedles to catalyze depropargylation reaction doxorubicin (DOX) prodrug imiquimod (IMQ) situ. The activated DOX subcutaneous induced released tumor-associated antigens. Concurrently, IMQ acts as a Toll-like receptor (TLR7) agonist, enhancing anti-tumor response. vivo experiments demonstrate that this achieves ≈97% inhibition tumors effectively inhibits untreated distant (≈94% inhibition) lung metastasis (≈92% inhibition).

The conventional molecular immunogenic cell death (ICD) inducers suffer from poor biocompatibility and unsatisfactory efficacy. Here, a biocompatible nanosized covalent organic framework (nCOF)–based pyroelectric catalyst (denoted as TPAD-COF NPs) is designed for catalysis-activated in situ immunotherapy. NPs confine molecules to rigid substantially reduce aggregation enhance biocompatibility, thus improving pyroelectrocatalytic efficiency. After tumor internalization, facilitate photothermal ablation under near-infrared (NIR) laser exposure, resulting effective ICD induction. In addition, effectively catalyze the conversion of temperature changes changes, which subsequently react with adjacent O 2 generate reactive oxygen species, triggering robust activation. vivo evaluation using mouse models confirmed that evidently inhibited proliferation primary distant tumors prevented lung metastasis NIR illumination. Therefore, this study opens an avenue designing nCOF-based catalysts
