Neurovascular unit (NVU) inflammation via activation of glial cells and neuronal damage plays a critical role in neurodegenerative diseases. Though the exact mechanism disease pathogenesis is not understood, certain biomarkers provide valuable insight into pathogenesis, severity, progression therapeutic efficacy. These markers can be used to assess pathophysiological status brain including neurons, astrocytes, microglia, oligodendrocytes, specialized microvascular endothelial cells, pericytes, NVU, blood-brain barrier (BBB) disruption. Damage or derangements tight junction (TJ), adherens (AdJ), gap (GJ) components BBB lead increased permeability neuroinflammation various disorders disorders. Thus, neuroinflammatory evaluated blood, cerebrospinal fluid (CSF), tissues determine neurological progression, responsiveness. Chronic common age-related Alzheimer's (AD), Parkinson's (PD), dementia. Neurotrauma/traumatic injury (TBI) also leads acute chronic responses. The expression some may altered many years even decades before onset In this review, we discuss neuroinflammation, neurodegeneration associated with disorders, especially those neurovascular pathologies. CSF, tissues. Neurofilament light (NfL), ubiquitin C-terminal hydrolase-L1 (UCHL1), fibrillary acidic protein (GFAP), Ionized calcium-binding adaptor molecule 1 (Iba-1), transmembrane 119 (TMEM119), aquaporin, endothelin-1, platelet-derived growth factor receptor beta (PDGFRβ) are important markers. Recent BBB-on-a-chip modeling offers promising potential for providing an in-depth understanding neurotherapeutics. Integration these clinical practice could potentially enhance early diagnosis, monitor improve outcomes.

Traumatic brain injury (TBI) is a complex condition that can resolve over time but all too often leads to persistent symptoms, and the risk of poor patient outcomes increases with aging. TBI damages neurons long axons within white matter tracts are critical for communication between regions; this causes slowed information processing neuronal circuit dysfunction. This review focuses on after multifactorial processes underlie damage, potential recovery, progression degeneration. A multiscale perspective across clinical preclinical advances presented encourage interdisciplinary insights from whole-brain neuroimaging down cellular molecular responses axons, myelin, glial cells tissue.

Background and aim Traumatic brain injury (TBI) poses a significant burden on the global economy due to its poor treatment prognosis. Current TBI markers do not comprehensively reflect disease status. Therefore, identifying more meaningful biomarkers is beneficial for improving prognosis clinical of patients. Methods The gene expression profile was obtained from Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) were subjected enrichment analysis, key potential identified through protein–protein interaction network cytoHubba modules. ROC curves used construct diagnostic models hub genes. Immunofluorescence experiments conducted detect candidate in rat models. Finally, we investigated normal human organs pan-cancer tumor tissues, evaluated their correlation with immune infiltration different tumors. Results A total 44 DEGs across four regions Enrichment analysis revealed that these primarily involved intracellular cell signal transduction pathways. Furthermore, three genes- RPS4Y1, KDM5D NLGN4Y-were module analysis. curve model also confirmed have high value serum. Subsequently, presence Kdm5d detected tissue rats immunofluorescence experiments. Compared rats, increased cortical area TBI no change hippocampus area, aligning observations Immune demonstrated changes subsets HIP PCx, revealing plasma cells CD8 T lowly (HIP) while neutrophils under-expressed (PCx). Pan-cancer indicated significantly up-regulated 23 cancers, down-regulated 3 associated 10 cancers. Conclusion Based results bioinformatics animal experiments, serves as biomarker diagnosis TBI. Additionally, research may develop into new serum markers, providing indicators further liquid biopsy aiding prevention both tumors certain extent.
