Onychomycosis caused by, e.g., Trichophyton rubrum or Candida albicans is the most common human nail disease with a worldwide prevalence of more than 10%. The therapeutic efficacy topical antimycotics for treatment onychomycosis proved to be inadequate in numerous studies on patients. main reasons are, above all, poor bioavailability active ingredients compartment, causing requirement extremely long application periods and correspondingly high demands adherence by patient. Abstract BACKGROUND Bacterial biofilm is a strong fortress for bacteria to resist harsh external environments, which can enhance their tolerance and exacerbate the drug/pesticide resistance risk. Currently, photopharmacology provides an advanced approach via precise spatiotemporal control regulating biological activities by light‐controlling molecular configurations, thereby having enormous potential in development of drug/pesticides. RESULTS To further expand application discovering new antibiofilm agents, we prepared series light‐controlled azo‐active molecules explored photo isomerization, fatigue resistance, anti‐biofilm performance. Furthermore, mechanisms inhibiting formation were systematically investigated. Overall, designed azo‐derivative A11 featured excellent anti‐ Xoo activity with half‐maximal effective concentration (EC 50 ) value 5.45 μg mL −1 , EC could be elevated 2.19 after ultraviolet irradiation (converted as cis ‐configuration). The photo‐switching behavior showed that had outstanding anti‐fatigue properties. An in‐depth analysis action mechanism effectively inhibit expression relevant virulence factors. This performance dynamically regulated loading private light‐switch property. CONCLUSION In this work, azo provide model resisting bacterial infection dynamic regulation formation. © 2024 Society Chemical Industry.

Imidazoles are crucial structural components in a variety of small-molecule inhibitors designed to target different kinases anticancer treatment. However, the effectiveness such is often hampered by nonspecific effects and development resistance. Photopharmacology provides compelling solution enabling external control over drug activity with spatiotemporal precision. Herein, we introduce novel strategy for caging bioactive triarylimidazole-based molecules. This approach involves introducing dialkylamino group as photoremovable on carbon atom imidazole ring, which intrinsically modulates core structure from planar tetrahedral 2H-imidazole, caged compound be selectively uncaged upon visible light exposure. We applied this innovative technique SB431542, inhibitor that targets pivotal TGF-β signaling pathway, dysregulation linked several human diseases, including cancer. Our results demonstrated selective inhibition breast cancer cell migration vitro activation, highlighting potential our transform molecules into light–activatable drugs, thereby facilitating regulation their pharmacological activity.

Multifunctional colloidal microgels that exhibit stimuli-responsive behaviour and excellent biocompatibility have attracted particular attention for developing functional compartmentalized networks. Herein, a series of (M0, M1, M2) were designed through the copolymerization di(ethylene glycol) methyl ether methacrylate (DEGMA) methacrylic acid (MAA) monomers using hydroxy ethyl methacrylate-coupled azobenzene (HEMA-Az) ethylene glycol dimetharylate (EGDMA) as crosslinkers. The in response to temperature, pH, light was thoroughly investigated spectroscopic, microscopic, light-scattering techniques. Interestingly, deswelled with an increase decrease under irradiation UV light. Such reversible swelling/deswelling exploited microgel M2, which showed better photoactuation at pH 5 higher fluid pumping velocity. actuating M2 optimized loading drug ciprofloxacin (Cf) study its release different conditions. Abstract This perspective article features the recent work published in this journal on functionalized azobenzene photoswitches, studied by Venkataramani and coworkers [Grewal, S. et al (2024) Photochem. Photobiol. , 1–16]. Azobenzene‐based photochromic molecules undergo reversible trans ‐to‐ cis isomerization under exposure to light. The reverse also takes place thermal conditions dark. study explores how different functional groups at various positions affect responses of these compounds light, their stabilities, isomeric ratios photostationary states, supramolecular behavior. Results show varied half‐lives, influenced terminal positions. Aggregation studies reveal formation architectures, from microcrystals gels with systems. Despite complexities large volume 30 compounds, research provides insights into designing efficient motifs.

Dibenzotriazonine represent a new class of nine-membered cyclic azobenzenes with nitrogen atom embedded in the bridging chain. To enable future applications this photoactive backbone, we propose study synthesis mono- and dihalogenated triazonines, that allow late-stage introduction different functionalized aryl groups heteroatoms (N, O, P) via palladium-catalyzed reactions. Indeed, diphenylphosphoryl-triazonines were synthesized functional such as aniline or phenol. Bis(diphenylphosphoryl)phenyl bis-carbamate-triazonines also isolated good yields.
