Abstract The bisbenzylisoquinoline alkaloids (bisBIAs) have attracted tremendous attention from the synthetic community due to their diverse and intriguing biological activities. Herein, we report convergent modular chemoenzymatic syntheses of eight bisBIAs bearing various substitutes linkages in 5–7 steps. gram‐scale synthesis well‐designed enantiopure benzylisoquinoline monomers was accomplished through an enzymatic stereoselective Pictet–Spengler reaction, followed by regioselective methylation or chemical functionalization a sequential one‐pot process. A modified intermolecular copper‐mediated Ullmann coupling enabled concise efficient total five different linear with either head‐to‐tail tail‐to‐tail linkage. biomimetic oxidative phenol dimerization selectively formed sterically hindered, electron‐rich diaryl ether bond, subsequent intramolecular Suzuki–Miyaura domino reaction facilitated first enantioselective three macrocyclic bisBIAs, including ent ‐isogranjine, tetrandrine O ‐methylrepandine. This study highlights great potential strategies paves way further explore functions these natural products.
