Abstract In 1966, Howard Roffwarg proposed the ontogenic sleep hypothesis, relating neural plasticity and development to rapid eye movement (REM) sleep, a hypothesis that current fetal neonatal research is still exploring. Recently, technological advances have enabled researchers automatically quantify architecture, which has caused resurgence of in this field as attempts are made further elucidate important role pre- postnatal brain development. This article will review our understanding driver identify possible areas for future research. Impact The evidence date suggests Roffwarg’s ontogenesis correct. A better relationship between functional connectivity needed. Reliable, non-invasive tools assess NICU at home need be tested real-world environment best way promote healthy needs understood before clinical trials promoting optimizing quality neonates could undertaken.

During the prenatal period, major brain development milestones are posed and calibrated through different mechanisms, among which endogenous activity, that prepares "system" to face external environment. However, specific nature of human nervous system, intended for plasticity is varied by area prolonged over time, requires much time environmental experiences shape cerebral circuitries. Therefore, neonate completely depends on caregiver, during first months postnatal life, it exhibits a transitory limited repertoire behaviors skills favors mother in her new role. This condition will gradually give way more mature competencies, within 2 years age. review takes perspective early attempts trace remarkable changes from in-utero period second year posing bridge between neurobiological substrate behavioral development. We based our work "normal" development, pointing out risks inherent any process.

Transient periods of childhood hearing loss can induce deficits in aural communication that persist long after auditory thresholds have returned to normal, reflecting long-lasting impairments the CNS. Here, we asked whether these behavioral could be reversed by treating one central impairments: reduction inhibitory strength. Male and female gerbils received bilateral earplugs a mild, reversible during critical period cortex development. After earplug removal return normal thresholds, trained tested animals on an amplitude modulation detection task. developmental induced both learning perceptual deficits, which were entirely corrected treatment with selective GABA reuptake inhibitor (SGRI). To explore mechanistic basis for findings, recorded amplitudes GABAA GABAB receptor-mediated IPSPs cortical thalamic brain slices. In loss-reared animals, IPSP significantly reduced within few days onset, this persisted into adulthood. SGRI prevented loss-induced amplitudes; but when administered period, it only restored amplitudes. These effects driven, part, ability upregulate α1 subunit-dependent responses. Similarly, ventral nucleus medial geniculate body. Thus, maintaining, or subsequently rescuing, GABAergic transmission thalamocortical pathway, some cognitive prevented.SIGNIFICANCE STATEMENT Even temporary normal. may arise from impairments, including synaptic inhibition. reinstating Gerbils reared transient displayed deficits.
