Mapping the endocytic vesicular acidification process is of prior importance to better understand health and pathological processes cells. Herein, by integrating a pH-sensitive i-motif pair fluorescence resonance energy transfer (FRET) into tetrahedral DNA framework (TDF), we develop pH-responsive nanomachine, allowing for efficient sensing pH from 7.0 5.5 via pH-triggered spatial proximity modulation FRET. The inheriting endo-lysosome-targeting ability TDF enables spatiotemporal tracking vesicle during endosomal maturation process. Analysis pH-dependent FRET response at single fluorescent spot level reveals significant difference between normal cancer performance nanomachine underlines its potential studies on acidification-related pathologies as universal platform.
