Abstract
Exosomes
are
extracellular
vesicles
that
actively
secreted
by
most
live
cells
via
endosome
pathway.
They
distribute
ubiquitously
in
body
fluids
and
play
important
roles
physiological
pathological
processes.
Recent
studies
have
revealed
exosomes
carry
cell‐specific
cargoes,
such
as
proteins
nucleic
acids,
which
been
recognized
emerging
noninvasive
biomarkers
liquid
biopsy.
Therefore,
the
molecular
profiling
of
tumor‐derived
is
crucial
facilitating
discovery
cancer‐related
biomarkers.
Recently,
various
techniques
proposed
for
sensitive,
accurate,
rapid
characterization
these
small
vesicles.
To
meet
growing
demands
multiplexed
detection,
microfluidics
has
integrated
with
diverse
traditional
analytical
methods
powerful
components
analysis.
Taking
advantages
their
inherent
merits
great
sensitivity,
high
analysis
performance
system
integration,
short
time,
low
cost,
volume
sample,
improved
potential
control,
automation,
disposability,
microfluidic
systems
significantly
exosomes.
In
this
review,
we
summarized
recent
progresses
technologies
exosomes,
including
proteins,
metabolites,
lipids.
Furthermore,
discussed
major
challenges
offered
a
perspective
strategy‐based
exosomal
discovery.
We
expected
in‐depth
summary
devices–assisted
will
greatly
facilitate
biomarkers,
thereby
realizing
highly
efficient
accurate
diagnosis,
prediction
cancers.
Moreover,
view
significance
interesting
approach
exosomes‐based
biopsy,
review
contribute
to
translation
technology
into
clinical
applications.

Exosomes

Exosomes,
a
subgroup
of
extracellular
vesicles
secreted
by
multiple
cells,
have
great
potential
as
cancer
biomarkers
in
clinical
applications.
However,
enrichment
and
detection
exosomes
from
complex
media
remain
huge
challenge
due
to
their
small
size.
Herein,
we
used
iodixanol
density
gradient
centrifugation
for
the
isolation
purification
label-free
exosomal
PD-L1
using
biochip
based
on
surface
plasmon
resonance
(SPR-ExoPD-L1).
The
obtained
are
lipid-bilayer
classical
exosome
markers
CD9,
CD63
CD81
highly
enriched.
Besides,
is
specifically
expressed
instead
non-vesicular
components
or
large
vesicles.
Compared
with
enzyme-linked
immunosorbent
assays,
SPR-ExoPD-L1
assay
could
better
distinguish
derived
melanoma
cells
different
levels
PD-L1.
Accurate
measurement
provide
critical
information
diagnosis
personalized
immunotherapy
cancer.
