Abstract N6-methyladenosine (m 6 A) is considered as the most common and important internal transcript modification in several diseases like type 2 diabetes, schizophrenia especially cancer. As a main target of m A methylation, long non-coding RNAs (lncRNAs) have been proved to regulate cellular processes at various levels, including epigenetic modification, transcriptional, post-transcriptional, translational post-translational regulation. Recently, accumulating evidence suggests that A-modified lncRNAs greatly participate tumorigenesis cancers. In this review, we systematically summarized biogenesis identified A-lncRNAs variety cancers, well their potential diagnostic therapeutic applications biomarkers targets, hoping shed light on novel strategies for cancer treatment.

Background: Long non-coding RNAs modulate tumor occurrence through different molecular mechanisms. It had been reported that HNF1A-AS1 (HNF1A Antisense RNA 1) was differently expressed in multiple tumors. The role of colorectal cancer less analyzed, and the mechanism regulating cell cycle has not completely elucidated. Methods: Differentially lncRNAs were screened out from TCGA database. examined CRC clinical samples lines by RT-qPCR. CCK8 assay, colony formation flow cytometry, transwell assays, tube forming assay vivo experiments performed to study function progression. Bioinformatic analysis, luciferase report pull-down RIP assays carried explore proteins binding potential downstream targets. Results: Our results showed upregulated associated with unfavorable prognosis. promoted proliferation, migration angiogenesis, accelerated reduced apoptosis CRC. Bioinformatics prediction further proved could promote CCND1 expression suppressing PDCD4 or competitively sponging miR-93-5p. Meanwhile, METTL3 mediated m6A modification affected its stability. HNF1A-AS1/IGF2BP2/CCND1 may act as a complex regulate stability CCND1. Conclusion: In summary, our result reveals novel which m6A-mediated axis promotes progression, along miR-93-5p upregulate CCND1, demonstrating significant regulation suggesting prognostic marker future.
