High-dimensional single-cell data has become an important tool in unraveling the complexity of immune system and its involvement homeostasis a large array pathologies. As technological tools are developed, researchers adopting them to answer increasingly complex biological questions. Up until recently, mass cytometry (MC) been main technology employed cytometric assays requiring more than 29 markers. Introduction Monitoring of innate myeloid cells (IMC) is broadly applied in basic and translational research, as well diagnostic patient care. Advances in cytometry have led to increases the number of cellular markers that are routinely measured. The resulting complexity data has prompted a shift from manual automated analysis methods. Currently, numerous unsupervised methods available cluster cells based on marker expression values. However, phenotyping clusters is typically not part process. Manually identifying both definitions (e.g. CD4 + , CCR7 CD45RA CD19 - ) and descriptive cell type names naïve T cells) values can be time-consuming, subjective, error-prone. In this work we propose an algorithm addresses these problems through creation tool, CytoPheno, assigns unidentified clusters. In the face of ever-present burden emerging and reemerging infectious diseases, there is a growing need to comprehensively assess individual- population-level immunity vaccine-preventable diseases (VPDs). Many these efforts, however, focus exclusively on antibody-mediated immunity, ignoring role T cells. Aimed at clinicians, public health practioners, others who play central roles in human vaccine research but do not have formal training immunology, we review how vaccines against elicit cell responses, what types responses are measured. We then use examples demonstrate six ways that cells contribute protection from VPD, including directly mediating protection, enabling antibody reducing disease severity, increasing cross-reactivity, improving durability, protecting special populations. conclude with discussion challenges solutions more widespread consideration clinical vaccinology.

While many algorithms for analyzing high-dimensional cytometry data have now been developed, the software implementations of these remain highly customized-this means that exploring a dataset requires users to learn unique, often poorly interoperable package syntaxes each step processing. To solve this problem, we developed {tidytof}, an open-source R using increasingly popular 'tidy data' interface.{tidytof} is available at https://github.com/keyes-timothy/tidytof and released under MIT license.
