Radiofrequency ablation (RFA), a thermoablative treatment for small hepatocellular carcinoma (HCC), has limited therapeutic benefit advanced HCC patients with large, multiple, and/or irregular tumors owing to incomplete RFA (iRFA) of the tumor mass. It is first identified that iRFA-treated exhibited increased pyruvate kinase M2 (PKM2) expression, exacerbated immunosuppression featured infiltration suppressive immune cells and proliferation, programmed cell death ligand 1 expression cancer ultimately poor prognosis. Herein, multifunctional nanomedicine fabricated by encapsulating nanoassemblies anti-PD-L1 spermidine-grafted oxidized dextran shikonin-containing lipid bilayers reverse iRFA-induced failure. Shikonin, PKM2 inhibitor, used suppress glycolysis in cells, while spermidine are introduced collectively reprogram proliferation functions infiltrated CD8+ T lymphocytes. Combined iRFA, which promoted exposure antigens, intravenous injection liposomal SPS-NPs effectively stimulated dendritic maturation reversed immunosuppression, thus eliciting potent antitumor immunity synergistically growth residual masses lung metastasis. The as-prepared promising potentiating benefits toward through reprogramming immunosuppression.

Abstract Photothermal therapy (PTT), as an innovative optical tumor treatment strategy, is characterized by its minimal invasiveness, high selectivity, long‐lasting efficacy, and safety, has attracted widespread attention. However, in order to effectively kill cancer cells, a higher local temperature very necessary, which inevitably results severe stress responses cell death, thereby triggering harmful inflammation. Additionally, achieving effective controlled release of drugs the microenvironment reduce systemic side effects still major challenge. Herein, intelligent nanoplatform first reported, featured sulfide iron (FeS) nanosheets core, loaded with perfluorohexane (PFH), skillfully coated polydopamine (PDA). It can serve generator reactive oxygen species (ROS) induce ferroptosis, further intensifying PTT, ROS eliminator normal tissue control undesired inflammatory response during differential strategy between surrounding tissue. Meanwhile, combination ultrasound‐targeted microbubble destruction (UTMD) technology significantly enhanced penetration drug. Overall, delivery FeS/PFH@PDA combined UTMD achieve precise efficient synergistic for tumors demonstrate outstanding ultrasound MR dual‐mode imaging performance.
