MicroRNAs (miRNAs) are short noncoding RNA sequences that regulate gene expression at the post-transcriptional level. They involved in regulation of multiple pathways, related to both physiological and pathological conditions, including autoimmune diseases, such as Systemic Sclerosis (SSc). Specifically, SSc is recognized a complex multifactorial disease, characterized by vascular abnormalities, immune dysfunction, progressive fibrosis, affecting skin internal organs. Among predisposing environmental triggers, evidence supports roles oxidative stress, chemical agents, viral infections, mostly those sustained beta-herpesviruses HCMV HHV-6. Dysregulated levels miRNA have been found patients compared healthy controls, intra- extracellular levels, providing sort signature disease. Notably, HCMV/HHV-6 infections were shown modulate profile, often superposing observed SSc, potentially promoting pathways associated with development. This review summarizes main data regarding alterations highlighting their potential prognostic or diagnostic markers for impact putative etiological agents on modulation.

Systemic lupus erythematosus (SLE) is a complex autoimmune disease marked by abnormal B-cell proliferation and increased autoantibodies. miRNAs play crucial role in regulating dysfunction SLE pathology. influence DNA methylation, activation, gene expression, contributing to pathogenesis. impact B cells through key processes like proliferation, differentiation, tolerance, apoptosis. also exacerbate inflammation immune responses modulating Interleukin 4 (IL-4), IL-6, interferon cytokines.
