Eosinophilic esophagitis (EoE) is a chronic, progressive, type 2 inflammatory disease with increasing global prevalence. An eosinophil-predominant inflammation that permeates the epithelium and deeper esophageal layers characterizes disease. Several cytokines, mainly derived from T-helper (Th2) cells epithelial cells, are involved in perpetuating responses by surface permeability promoting tissue remodeling characterized epithelial–mesenchymal transition (EMT) collagen deposition. This leads to strictures narrow caliber esophagi, which proportional patient’s age untreated length. Pathophysiological mechanisms leading EoE have been described recent years, transforming growth factor beta (TGF)-beta fibrotic phenomena EoE. However, evidence on dependence of these TGF-beta scarce contradictory. review provides state-of-the art knowledge intimate fibrosis its clinical consequences.

Abstract Background Radiotherapy is the main treatment modality for thoracic tumours, but it may induce pulmonary fibrosis. Currently, pathogenesis of radiation‐induced fibrosis (RIPF) unclear, and effective treatments are lacking. Transforming growth factor beta 1 (TGFβ1) plays a central role in RIPF. We found that activated TGFβ1 had better performance radiation pneumonitis (RP) risk prediction by detecting total levels patient serum. αv integrin key roles activation, integrin‐mediated activation RIPF unclear. Here, we investigated application antagonist cilengitide to prevent Methods Itgav loxP/loxP ;Pdgfrb‐Cre mice were generated conditionally knocking out myofibroblasts, wild‐type treated with or placebo. All received 16 Gy underwent sham procedure. Lung was measured modified Ashcroft score microcomputed tomography (CT). An enzyme‐linked immunosorbent assay (ELISA) used measure serum concentration, Smad2/3 p‐Smad2/3 determined via Western blotting. Results Conditional knockout significantly attenuated ( p < .01). Hounsfield units (HUs) lungs reduced compared control .001). decreased active secretion inhibited fibroblast expression. Exogenous TGFβ1, not latent reversed these reductions. Furthermore, elicited similar results prevented Conclusions The present study revealed conditional both inhibiting activation. Highlights Activated has superior capacity predicting vital development (RIPF). knock myofibroblasts from developing Cilengitide alleviated be targeted approaches preventing

Inhibitors of the transforming growth factor-β (TGF-β) pathway are potentially promising antifibrotic therapies, but nonselective simultaneous inhibition all three TGF-β homologs has safety liabilities. TGF-β1 is noncovalently bound to a latency-associated peptide that is, in turn, covalently different presenting molecules within large latent complexes. The TGF-β-binding proteins (LTBPs) present extracellular matrix, and presented on immune cells by two transmembrane proteins, glycoprotein A repetitions predominant (GARP) leucine-rich repeat protein 33 (LRRC33). Here, we describe LTBP-49247, an antibody selectively inhibited activation LTBPs did not bind GARP or LRRC33. Structural studies demonstrated LTBP-49247 recognized epitope LTBP-presented accessible GARP- LRRC33-presented TGF-β1, explaining antibody's selectivity for LTBP-complexed TGF-β1. In rodent models kidney fibrosis etiologies, attenuated fibrotic progression, indicating central role renal fibrosis. mice, have toxic effects associated with less selective inhibitors. These results establish feasibility targeting LTBP-bound as approach treating
