Abstract Anticancer immunotherapy is hampered by the poor tumor immunogenicity and immunosuppressive microenvironment (TME). Herein, a liposome nanodrug co‐encapsulating doxycycline hydrochloride (Doxy) chlorin e6 (Ce6) to simultaneously induce autophagy inhibition mitochondria dysfunction for potentiating photo‐immunotherapy developed. Under near infrared laser irradiation, Ce6 generates cytotoxic reactive oxygen species (ROS) elicits robust photodynamic therapy (PDT)‐induced immunogenic cell death (ICD) TME remodeling. In addition, Doxy induced dysfunction, which increases ROS generation enhances PDT exert more potent killing effect powerful ICD. Meanwhile, MHC‐I expression on cells surface efficient inhibition, leading antigen presentation CTLs recognition increase immunogenicity. The nanodrugs elicit remarkable antitumor combining Ce6‐mediated Doxy‐induced dysfunction. developed represent highly strategy improving cancer immunotherapy.
