Sonodynamic therapy (SDT) has promising application prospects in tumor therapy. However, SDT does not eradicate metastatic tumors. Herein, Cu-substituted ZnAl ternary layered double hydroxide nanosheets (ZCA NSs) were developed as both sonosensitizers and copper nanocarriers for synergistic SDT/cuproptosis cancer An optimized electronic structure more conducive to the sonodynamic process was obtained from ZCA NSs via Jahn–Teller effect induced by introduction of Cu2+, synthesized regulated intricate microenvironment (TME) depleting endogenous glutathione (GSH) amplify oxidative stress further enhanced performance. Furthermore, cuproptosis evoked intracellular overload Cu2+ amplified SDT, leading irreversible proteotoxicity. In vitro results showed that such synergetic triggered immunogenic cell death (ICD) promoted maturation dendritic cells (DCs). as-synthesized NS-mediated thoroughly eradicated vivo solid tumors simultaneously elicited antitumor immunity suppress lung liver metastasis. Overall, this work established a nanoplatform with satisfactory immunity.

Abstract Copper is a necessary micronutrient for maintaining the well-being of human body. The biological activity organic ligands, especially their anticancer activity, often enhanced when they coordinate with copper(I) and (II) ions. its compounds are capable inducing tumor cell death through various mechanisms action, including activation apoptosis signaling pathways by reactive oxygen species (ROS), inhibition angiogenesis, induction cuproptosis, paraptosis. Some copper complexes currently being evaluated in clinical trials ability to map hypoxia cancers, locally advanced rectal cancer bulky tumors. Several studies have shown that nanoparticles can be used as effective agents chemodynamic therapy, phototherapy, hyperthermia, immunotherapy. Eight 2,2′:6′,2″-terpyridines, substituted at the 4′-position with aromatic groups featuring variations in π-conjugation, ring size, heteroatoms, and methoxy groups, were employed to enhance antiproliferative potential of [Cu2Cl2(R-terpy)2](PF6)2. Assessing cytotoxicity A2780 (ovarian carcinoma), HCT116 (colorectal HCT116DoxR carcinoma resistant doxorubicin) normal primary fibroblasts revealed that Cu(II) complexes 4-quinolinyl, 4-methoxy-1-naphthyl, 2-furanyl, 2-pyridynyl substituents showed superior therapeutic cells significantly reduced (42–129× lower). Besides their cytotoxicity, are able increase intracellular ROS interfere cell cycle progression, leading death by apoptosis autophagy. Importantly, they demonstrated antimetastatic antiangiogenic properties without vivo toxicity. Accurate detection of copper ions is crucial since it an essential trace element. Carbon quantum dots (CQDs) were prepared from apricot leaves using a hydrothermal method. These CQDs, which generated reference signal, combined with glutathione-bound gold nanoclusters (GSH-AuNCs) synthesized by the reduction chloroauric acid to form ratiometric fluorescent probe. The probe exhibited fluorescence emission at 450 nm and 568 nm. determination Cu(II) was performed through visual analysis color change intensity ratio (I450/I568) peaks. Orange GSH-AuNCs responded fluorescence, blue CQDs chemically inert. probes changed pink purple in response different concentrations Cu(II). linear range for 0 120 µmol/L, limit (LOD) 0.65 µmol/L. This study has demonstrated that provided good selectivity detecting water potential practical application.
