Astragaloside IV (AS-IV), a major bioactive component of Astragalus membranaceus, exhibits anti-cancer and anti-inflammatory properties. However, its precise role in nasopharyngeal carcinoma (NPC) remains unclear. This study investigated the effects AS-IV on NPC progression relationship with Special AT-rich binding protein-2 (SATB2), diagnostic marker for NPC. treatment reduced cell viability dose-dependent manner, as assessed by CCK-8 assays. Functional experiments, including transwell, immunofluorescence, flow cytometry assays, demonstrated that inhibited migration, invasion, autophagy while promoting apoptosis. Western blot analysis showed SATB2 expression was significantly elevated cells, particularly C666-1 HK-1 cells. Overexpression partially reversed AS-IV's inhibitory progression. Further revealed suppressed Wnt signaling pathway downregulating expression, overexpression restored activation. effect upon inhibitor DKK-1. In vivo, administration tumor growth nude mouse subcutaneous xenograft model, Ki-67 positivity, lowered LC3B indicating decreased proliferation autophagy. these were diminished overexpression. These findings suggest exerts anti-tumor suppressing activation, highlighting potential therapeutic agent (AS-IV) reduces vitality, suppresses invasion autophagy, fosters apoptosis.SATB2 notably high levels cells.Overexpression counteracts inhibition malignant AS-IV.AS-IV impedes decreasing thereby hindering pathway.AS-IV deters mice.

The development and progression of myopia are influenced by the Wnt7b/β-catenin signaling pathway. This study investigated specific impacts this pathway on biomechanical properties sclera altering expression matrix metalloproteinase-2 (MMP-2) to regulate type I collagen (collagen I) levels. We examined effects MMP-2 human fetal scleral fibroblasts (HFSFs) guinea pigs with form-deprivation (FDM). To explore role in context, we treated HFSFs pig agonists inhibitors targeting MMP-2. levels Wnt7b, MMP-2, were subsequently analyzed quantitatively via western blot (WB) analysis, immunofluorescence, quantitative real-time PCR (qRT-PCR) assess protein mRNA changes response manipulation. Atomic force microscopy (AFM) was used measure elastic modulus directly evaluate cell tissue stiffness. In FDM model, essential ocular parameters such as refractive error axial length (AL) also assessed. vivo vitro activation significantly upregulated expression, which accompanied a notable decrease change led reduction both FDM. These significant indicated Alterations biomechanics associated parameters, including an increase AL myopic shift refraction. regulates upregulating leads increased degradation and, consequently, elongation error.
