Quercetin, a prevalent flavonol compound, has gained attention for its multifaceted mechanisms of action against various cancers, highlighting potential as an adjunctive therapy in cancer treatments. This review aims to systematically evaluate the structural optimization, action, and clinical applications quercetin nano-derivatives treatment. Employing bibliometric analysis 6231 articles from Web Science Core Collection, we observed notable increase annual publications, particularly USA China, indicating growing interest quercetin's therapeutic potential. Our findings reveal that enhances efficacy conventional therapies by modulating critical signaling pathways, thereby increasing cell sensitivity while simultaneously protecting normal tissues therapy-induced damage. Structural modifications, including glycosylation, methylation, sulfation, glucuronidation, alongside nanoparticle formulation, significantly improve stability, solubility, bioavailability quercetin, enabling targeted drug delivery. Despite promising preclinical outcomes, translation remains nascent, necessitating further rigorous research validate safety human subjects. In conclusion, exhibits substantial anticancer properties potential, future studies should focus on expanding sample sizes, elucidating metabolic conducting comprehensive trials inform application oncology.
