Abstract Lipidomics plays an essential role in the development of improved understanding lipids metabolism and identification new biomarkers or therapeutic targets related diseases. The strong analytical power mass spectrometry its rapid developments respect instruments techniques have significantly accelerated emerging lipidomics application fields biology, medicine, pharmacy. strategy chemical derivatization can remarkably improve shortcomings spectrometric technologies shotgun liquid chromatography spectrometry, past decade many studies been reported for fatty acids, glycerophospholipids, sphingomyelins, monoglycerides, diacylglycerols, long‐chain bases, steroids, so on. Therefore, this review will focus on approaches about research progresses shotgun‐based spectrometry–based targeted (from 2005 to July 2019, most reports emerged 5 years), put forward problems prospects field. It is expected be helpful design synthesis reagents, especially outstanding stable isotope labeling strategies matched analysis methods.

Endocannabinoids (eCBs) are endogenous lipid messengers that primarily bind cannabinoid receptors CB1/CB2 and together with the enzymes regulate their biosynthesis degradation define endocannabinoid system. The eCB signaling system plays a key role in central nervous system, results often altered neurological disorders. analysis of eCBs is challenging due to low concentration biospecimens, this exacerbated Mass Spectrometry Imaging (MSI) where sensitivity tissue dependent ion suppression obscure spatial visualization. In work we address limitation by application laser-induced post-ionization (MALDI-2) approach. Herein demonstrate MALDI-2 boosts detection 2-arachidonylglycerol (2-AG) N-acylethanolamines (AEA, PEA, OEA) respect MALDI, can be visualized brain at physiological only MALDI-2-MSI. Root-mean-square (RMS), Total count (TIC) internal standards (I.S.) normalization were evaluated, I.S. providing improved pixel variation more uniform distribution for 2-AG PEA specific regions. Furthermore, high resolution up 5 μm size was resulting all confirming potential even reducing ablated amount. As proof concept, method applied map mouse model mild traumatic injury, APP-SWE mice, highlighting differences modulation Cortex, Hippocampus Hypothalamus, suggesting ability reveal valuable biological insights neuropharmacology.

ABSTRACT Quantitative lipidomic analysis performed by mass spectrometry is required for determination of the lipid content extracellular vesicles (EVs). Such methods can provide information about total amount lipids, species composition, purity EV samples as well cellular origin EVs. There are, however, many pitfalls when performing analyses. Thus, any non‐specialist should collaborate with experts in lipidomics. In addition to good review articles giving advice analyses, we recommend and guidelines published Lipidomic Standard Initiative, an interest group affiliated International Lipidomics Society.

Lipids and metabolites are of interest in many clinical research settings because it is the metabolome that increasingly recognized as a more dynamic sensitive molecular measure phenotype. The enormous diversity lipid structures importance biological structure-function relationships wide variety applications makes accurate identification challenging yet crucial area community. Indeed, subtle differences chemical lipids can have important implications cellular metabolism disease pathologies. speed, sensitivity, specificity afforded by modern mass spectrometry has led to its widespread adoption field lipidomics on different instrument platforms experimental workflows. However, unambiguous complete structural remains challenging. Increasingly sophisticated tandem (MS/MS) approaches now being developed seamlessly integrated into workflows meet this challenge.
