This PhD thesis, entitled "Holographic biosensors made of DNA-functionalised hydrogels for in vitro diagnostics", focuses on the development an analyte-sensitive hydrogel, functionalised with oligonucletide probes, a diffractive structure as optical transducer diagnostic applications.The first chapter includes overview different concepts related to biosensing, recent developments diagnostics market and, particular, DNA biosensors.In addition, synthesis and characterization hydrogels, their role support matrix immobilization strategies are presented.Finally, basic holography new detection strategy diffraction gratings biosensing explained.Next, Chapter 2, objectives this project discussed.The aim research is LIST OF PUBBLICATIONS

Gene therapy can potentially treat a great number of diseases, from cancer to rare genetic disorders. Very recently, the development and emergency approval nucleic acid-based COVID-19 vaccines confirmed its strength versatility. However, gene encounters limitations due lack suitable carriers vectorize therapeutic material inside target cells. Nanogels are highly hydrated nano-size crosslinked polymeric networks that have been used in many biomedical applications, drug delivery tissue engineering diagnostics. Due their easy production, tunability, swelling properties they called attention as promising vectors for delivery. In this review, nanogels discussed acid aiming enlarge therapy's window. Recent works highlighting optimization inherent transfection efficiency biocompatibility reviewed here. Abstract Polyhedral oligomeric silsesquioxane (POSS) is a 3D, cage‐like nanoparticle with an inorganic Si–O–Si core and eight tunable corner functional groups. Its well‐defined structure grants it distinctive physical, chemical, biological properties has been widely used for preparing high‐performance materials. Recently, click chemistry enabled the synthesis of various POSS‐based materials diverse biomedical applications. This article reviews recent applications in field, including cancer treatment, tissue engineering, antibacterial use, imaging. Representative examples are discussed detail. Among applications, treatment engineering most important. Finally, this review presents current limitations provides guidance future research.

Both azido (N3) and trifluoromethyl (CF3) groups are key moieties of numerous valuable molecules that extensively applied in drug discovery, chemical biology, synthetic chemistry. However, the asymmetric construction chiral quaternary stereocenters bearing both N3 CF3 is still unexplored. Herein, we report a kind bench-stable easily adjustable benziodazolone-based azidating reagents. These reagents were used to achieve an enantioselective copper-catalyzed azidation N-unprotected 3-trifluoromethylated oxindoles provide diverse enantioenriched 3-N3-3-CF3 oxindoles.
