Oxidative stress and AKT serine-threonine kinase (AKT) are responsible for regulating several cell functions of cancer cells. Several natural products modulate both oxidative anticancer effects. However, the impact product-modulating on lacks systemic understanding. Notably, contribution by downstream effectors is not yet well integrated. This review explores role pathway (AKT/AKT effectors) ten functions, including apoptosis, autophagy, endoplasmic reticulum stress, mitochondrial morphogenesis, ferroptosis, necroptosis, DNA damage response, senescence, migration, cell-cycle progression. The connected to these through function mediators. Moreover, related Based this rationale, with modulating abilities exhibit potential regulate but some were rarely reported, particularly effectors. sheds light understanding roles in providing future directions treatment.

Regulated cell death (RCD), also well-known as programmed (PCD), refers to the form of that can be regulated by a variety biomacromolecules, which is distinctive from accidental (ACD). Accumulating evidence has revealed RCD subroutines are key features tumorigenesis, may ultimately lead establishment different potential therapeutic strategies. Hitherto, targeting with pharmacological small-molecule compounds been emerging promising avenue, rapidly progressed in many types human cancers. Thus, this review, we focus on summarizing not only apoptotic and autophagy-dependent signaling pathways, but crucial pathways other subroutines, including necroptosis, pyroptosis, ferroptosis, parthanatos, entosis, NETosis lysosome-dependent (LCD) cancer. Moreover, further discuss current situation several improve cancer treatment, such single-target, dual or multiple-target compounds, drug combinations, some new strategies would together shed light future directions attack vulnerabilities drugs for purposes.

Mitochondria are dynamic organelles that have essential metabolic activity and regarded as signalling hubs with biosynthetic, bioenergetics functions orchestrate key biological pathways. However, mitochondria can influence all processes linked to oncogenesis, starting from malignant transformation metastatic dissemination. In this review, we describe how alterations in the mitochondrial status contribute acquisition of typical traits, discussing most recent discoveries many unanswered questions. We also highlight expanding our understanding regulation function mechanisms context cancer cell metabolism could be an important task biomedical research, thus offering possibility targeting for treatment cancer.

Abstract Triple-negative breast cancer (TNBC) is a subtype of human with one the worst prognoses, no targeted therapeutic strategies currently available. Regulated cell death (RCD), also known as programmed (PCD), has been widely reported to have numerous links progression and therapy many types cancer. Of note, RCD can be divided into different subroutines, including autophagy-dependent death, apoptosis, mitotic catastrophe, necroptosis, ferroptosis, pyroptosis anoikis. More recently, targeting subroutines small-molecule compounds emerging promising strategy, which rapidly progressed in treatment TNBC. Therefore, this review, we focus on summarizing molecular mechanisms above-mentioned seven major related TNBC latest progress subroutines. Moreover, further discuss combined drug (e.g., narciclasine) or more drugs torin-1 chloroquine) achieve potential by regulating importantly, demonstrate several ONC201 NCT03733119) clinical trials. Taken together, these findings will provide clue illuminating actionable low-hanging-fruit druggable targets candidate for RCD-related therapies. Graphical abstract

Patients with hypertension have a risk of depression. Morinda officinalis oligosaccharides (MOOs) anti-depressant properties. In this study, we aimed to determine whether MOOs can improve the symptoms depression in individuals hypertension.Dahl salt-sensitive rats fed high-salt diet were stimulated by chronic unpredictable mild stress mimic Primary astrocytes and neurons isolated from these rats. Astrocytes underwent LPS stimulation simulate inflammatory during administrated at 0.1 mg/g/day vivo 1.25, 2.5, 5 mg/mL vitro. Mitophagy was inhibited using mM 3-methyladenine (3-MA). Abstract Hepatocellular carcinoma (HCC) is the most common primary liver cancer with a high mortality rate. It regarded as significant public health issue because of its complicated pathophysiology, metastasis, and recurrence rates. There are no obvious symptoms in early stage HCC, which often leads to delays diagnosis. Traditional treatment methods such surgical resection, radiotherapy, chemotherapy, interventional therapies have limited therapeutic effects for HCC patients or metastasis. With development molecular biology immunology, signaling pathways immune checkpoint were identified main mechanism progression. Targeting these molecules has become new direction HCC. At present, combination targeted drugs inhibitors first choice advanced patients. In this review, we mainly focus on cutting‐edge research corresponding therapy immunotherapy great significance comprehensively understand pathogenesis search potential targets, optimize strategies
