Although the first discovery of a non-coding RNA (ncRNA) dates back to 1958, only in recent years has complexity transcriptome started be elucidated. However, its components are still under investigation and their identification is one challenges that scientists presently facing. In addition, function far from being fully understood. The portion genome indeed largest, both quantitatively qualitatively. A large fraction these ncRNAs have regulatory role either coding mRNAs or other ncRNAs, creating an intracellular network crossed interactions (competing endogenous networks, ceRNET) fine-tune gene expression health disease. alteration equilibrium among such can enough cause transition disease, but opposite equally true, leading possibility intervening based on mechanisms cure human conditions. this review, we summarize present knowledge mechanisms, illustrating how they used for disease treatment, current pitfalls, roles environmental lifestyle-related contributing factors, addition ethical, legal, social issues arising (improper) use.

Research over the past two decades has confirmed that noncoding RNAs (ncRNAs), which are abundant in cells from yeast to vertebrates, no longer “junk” transcripts but functional regulators can mediate various cellular and physiological processes. The dysregulation of ncRNAs is closely related imbalance homeostasis occurrence development diseases. In mammals, ncRNAs, such as long (lncRNAs) microRNAs (miRNAs), have been shown serve biomarkers intervention targets growth, development, immunity, disease progression. regulatory functions lncRNAs on gene expression usually mediated by crosstalk with miRNAs. most predominant mode lncRNA-miRNA lncRNA-miRNA-mRNA axis, act competing endogenous (ceRNAs). Glioblastoma multiforme (GBM)is a highly aggressive malignancy of the central nervous system characterized by poor survival rates. Ferroptosis, an iron-dependent cell death pathway, is promising therapeutic target for GBM. However, current treatments targeting pathways have not yielded expected results. Long noncoding RNAs (lncRNAs) been implicated in tumour proliferation, however, their role ferroptosis GBM remains underexplored. This study investigated interplay between lncRNA LINC01088 and to identify novel strategies. We conducted gain- loss-of-function studies assess impact on tumourigenesis both vitro vivo. Bioinformatics, dual-luciferase reporter assays, chromatin immunoprecipitation, RNA pulldown, mass spectrometry, immunoprecipitation (RIP), transcriptome sequencing were utilized elucidate mechanisms underlying expression its downstream effects ferroptosis. Research indicates that LINC00707 is abnormally expressed in patients suffering from diabetic kidney disease (DKD). Nevertheless, the precise role of context DKD remains enigmatic and warrants further investigation. Initially, RT-qPCR was employed to assess expression levels with DKD. Subsequently, we cultured glomerular podocytes under high-glucose conditions. The effects on cell proliferation, apoptosis, podocyte functionality (Podocin, Nephrin, CD2AP), oxidative stress (MDA SOD), inflammatory responses (IL-1β, IL-6, TNF-α) were evaluated using CCK-8, flow cytometry, RT-qPCR, ELISA assays. Finally, co-regulated alongside its downstream targets elucidate molecular mechanisms by which influences injury. upregulated patients. Furthermore, a notable inverse correlation has been observed between renal function these Upon downregulating LINC00707, an increase cellular proliferative activity, elevated Podocin, CD2AP. Concurrently, reduction associated decreased MDA, as well proinflammatory cytokines. Significantly, inhibition miR-223-3p found reverse effects. Further investigation revealed directly FK506-binding protein 5 (FKBP5). Inhibition may reduce damage during hyperglycemia targeting miR-223-3p/FKBP5.
