Scrophularia ningpoensis, a perennial medicinal plant from the Scrophulariaceae family, is original species of Scrophulariae Radix (SR) in Chinese Pharmacopoeia. This medicine usually deliberately substituted or accidentally contaminated with other closely related including S. kakudensis, buergeriana, and yoshimurae. Given ambiguous identification germplasm complex evolutionary relationships within genus, complete chloroplast genomes four mentioned were sequenced characterized. Comparative genomic studies revealed high degree conservation structure, gene arrangement, content species, entire genome spanning 153,016-153,631 bp full length, encoding 132 genes, 80 protein-coding 4 rRNA 30 tRNA 18 duplicated genes. We identified 8 highly variable plastid regions 39-44 SSRs as potential molecular markers for further genus. The consistent robust phylogenetic ningpoensis its common adulterants firstly established using total 28 family. In monophyletic group, kakudensis was determined to be earliest diverging succeeded by ningpoensis. Meanwhile, yoshimurae buergeriana clustered together sister clades. Our research manifestly illustrates efficacy identifying counterfeits will also contribute deeper understanding processes Scrophularia.

Biodiversity underlies ecosystem resilience, function, sustainable economies, and human well-being. Understanding how biodiversity sustains ecosystems under anthropogenic stressors global environmental change will require new ways of deriving applying data. A major challenge is that data knowledge are scattered, biased, collected with numerous methods, stored in inconsistent ways. The Group on Earth Observations Observation Network (GEO BON) has developed the Essential Variables (EBVs) as fundamental metrics to help aggregate, harmonize, interpret observation from diverse sources. The plant adaptation response to a changing environment depends on the genetic diversity level it possesses. Genetic and thorough understanding of population indices are pivotal for decoding dynamic environmental stressors. development polymerase chain reaction (PCR)-based molecular markers enables comprehensive analyses precise detection individuals groups with unique variations. Various have been employed assess diversity, examine structure, delineate cluster patterns within among populations. DNA revolutionize studies by allowing detailed variations, including economically significant trait-influencing genes. Despite their simplicity, they offer high reproducibility, ensuring accurate estimations variation. Integrating multiple marker systems advanced high-throughput sequencing techniques is poised enhance management depleting resources providing picture at genome-wide level. This review explores diverse markers, elucidating advantages limitations, highlights impact evaluating structure plants.

Present review describes the molecular tools and strategies deployed in trait discovery improvement of major crops. The prospects challenges associated with these approaches are discussed. Crop relies on modulating genes genomic regions underlying key traits, either directly or indirectly. Direct include overexpression, RNA interference, genome editing, etc., while breeding majorly constitutes indirect approach. With advent latest technologies, could hasten crop species. Next-generation sequencing, high-throughput genotyping, precision use space technology for accelerated growth, etc. had provided a new dimension to programmes that work towards delivering better varieties cope up challenges. Also, studies have widened from understanding response plants single stress combined stress, which provides insights into mechanisms regulating tolerance more than one at given point time. Altogether, next-generation genetics genomics made tremendous progress improved varieties; however, scope still exists expand its horizon other species remain underutilized. In this context, present systematically analyses different serve as roadmap executing similar application, pros, cons, each approach been discussed examples, altogether, comprehensive coverage advances meet ever-growing demands agricultural produce.
