A novel primer (FaSt-R) targeting the Prunus-specific Falling Stone (FaSt) non-autonomous transposon was combined with start-codon-targeted (SCoT) primers to assess genetic diversity in 12 cultivars from six Prunus species and 28 of European plum. Compared SCoT-only analyses, SCoT–FaSt combination produced fewer total bands but a higher percentage polymorphic bands, while maintaining comparable values for polymorphism information content, resolving power, gene diversity, Shannon’s index. markers generated across broader size range, which made gel patterns less dense, enabling more accurate detection differentially amplified fragments. Neighbor-joining principal component analyses confirmed that provided sufficient phylogenetic resolution at both interspecific intraspecific levels. The sequencing 32 amplicons revealed FaSt elements 26 fragments, SCoT preferentially annealing GC-rich exonic intergenic regions. Seventeen protein-coding one RNA-coding were partially identified, localized UTRs introns genes key physiological functions. Comparative analysis indicated biased distribution between Cerasus subgenera. In silico findings suggest are fragmented cherry species, potentially contributing subgeneric divergence. Overall, marker system is effective evaluating reconstructing relationships, elucidating genomic impact an active Mutator-like transposon.

Micropropagation has an important role in the large-scale production of blackberry plant material, given high proliferation rates this species. The aim present study was to evaluate proliferative capacity grown vitro on wheat starch-gelled culture medium compared classical agar-gelled and assess genetic fidelity between proliferated shoots their mother plants. Six varieties (‘Čačanska Bestrna’, ‘Chester Thornless’, ‘Driscoll’s Victoria’, ‘Loch Ness’, ‘Polar’, ‘Karaka Black’) were tested. For proliferation, Murashige Skoog (MS) supplemented with 0.5 mg dm−3 6-benzyladenine (BA) used. conventional gelled 0.5% agar, starch used as alternative gelling agent a concentration 5%. results showed that for all cultivars, highest number shoots/inoculum obtained medium, maximum value 54.42 ± 4.18 presented by Black’. Considering length shoots, tested cultivars outstanding 5% starch. values regarding observed Thornless’ followed ‘Čačanska Ness’ 5.55 0.04 cm, 5.46 0.06 5.37 0.09 respectively. uniformity micropropagated relation plants confirmed sequence-related amplified polymorphism (SRAP) start codon targeted (SCoT) molecular markers.

Juglans regia L. is an edible fruit tree cultivated worldwide for its fruits and wood as urban tree. Globally, there growing concern preserving the genetic diversity of trees with high economic ecological value. This study investigates J. in landscapes assesses quality walnut kernels a local food product. An inventory 150 from five populations public green spaces Lugoj, Caransebeș Jupa, well two semi-natural hilly ecosystems Banat Region, was conducted. Molecular analyses showed that Directed Amplification Minisatellite-region DNA (DAMD) markers were more effective than Start Codon Targeted (SCoT) markers, higher average polymorphism 56.26%, compared to 49.44%. DAMD07 achieved 100% DAMD05 strong balance between P.I.C. (0.35) (54.54%). Chemical analysis revealed following contents kernels: protein (12.81% 16.80%), lipids (60.39% 69.08%), total polyphenols (5484.66 10,788.4 mg GAE/kg), copper (3.655 8.532 mg/kg), manganese (14.408 28.618 zinc (19.813 46.583 lead (1.204 2.27 mg/kg) cadmium (0.03451 0.08065 mg/kg). These findings are critical conservation efforts, forestry management ensuring safety products derived regia.

Pot marigold is an ornamental plant of great importance in pharmacy and cosmetology. However, there limited published information on the diversity species at both morphological genetic levels. This paper aimed to determine selected varieties. The research focused varieties with different flower colours, most important features flowers height were analysed. Genetic analyses performed using start codon target (SCoT) markers. correlation coefficients between analysed studied showed a significant positive relationship height, number inflorescences per inflorescence diameter. grouped according colour their flowers. results obtained our work indicate that, despite variability observed features, gene pool limited. can be valuable for breeders, particularly selecting suitable crossbreeding breeding programs. The analysis was conducted to assess the inter-varietal genetic polymorphism of Poa pratensis based on PCR technology using nine SCoT markers. informative primers for variety differentiation have been identified; parameters diversity were estimated: Shannon’s index, Nei’s effective number alleles and indicator information content (PIC — content). Genetic structure analyzed collection evaluated Neighbor-Joining dendrogram PCoA-analysis.
