Abstract Background The greater omentum comprises peritoneal, adipose, vascular, and lymphoid tissues. Most omental malignancies are metastatic tumors, the incidence of primary tumors is rare. We report on a prior smooth muscle tumor case in an adult male patient. Case presentation A 54-year-old Japanese patient with no relevant medical history was diagnosed abdominal mass during routine checkup. Subsequent contrast-enhanced computed tomography revealed approximately 3 cm size omentum, laparotomy performed. 27 × 25 20 mm raised lesion found omentum. Microscopically, spindle cells were observed arranged whorls fascicles. Individual had short spindle-shaped nuclei slightly increased chromatin characterized by eosinophilic, cytoplasm. mitotic count less than 1 per 50 high-power fields. showed positive immunoreactivity for α actin, HHF35, desmin immunohistochemical examination. Ki-67 labeling index using average method 1.76% (261/14806). No any other tested markers. considered leiomyoma owing to lack malignant findings. However, has rarely been reported, it can be difficult completely rule out potential soft Our decisively considering leiomyoma. Consequently, did not undergo additional adjuvant therapy followed up. satisfied treatment neither recurrence nor metastasis at 13-month postoperative follow-up. Abstract Formalin-fixed paraffin-embedded (FFPE) tissue samples are important for genomic analysis of thyroid carcinomas, particularly various molecularly targeted therapies. Therefore, this study developed and validated a technique preparing FFPE that preserves nucleic acid quality, which is fundamental precise analysis, more effectively than conventional methods. We analyzed surgically resected gland tumors, lymph node metastases, separately fixed tumor to optimize formalin fixation assess the influence specimen type preparation methods on quality. assessed several quality indicators, including DNA integrity number, cycle threshold ratio, RNA DV200. Formalin-fixed paraffin-embedded (FFPE) tissue samples are widely used for genomic analyses in clinical and research settings. However, prolonged storage at room temperature may compromise nucleic acid quality, potentially affecting the reliability of molecular testing. In this study, we investigated impact various temperatures repeated changes on quality FFPE samples. Four surgically resected tumor cases were analyzed. Following cold ischemia times 29-60 min, tissues sampled upon arrival pathology department, divided into five equal parts, fixed 10 % neutral buffered formalin. The resulting blocks stored 18 °C, 4 -20 -80 or - 150 °C. After storage, returned to sectioning analysis. DNA RNA assessed 3, 6, 12 months using integrity number (DIN), short-to-long cycle threshold (S/L Ct) ratio, (RIN), DV200. Samples °C exhibited time-dependent deterioration certain indicators, whereas those below maintained stable quality. Despite multiple freeze-thaw cycles, no significant degradation was observed under colder conditions. Our findings suggest that is a feasible effective approach preserving tissues.
