Contaminated food with Shiga-toxin producing Escherichia coli (STEC) can cause diarrhea and severe diseases in consumers. This study aimed to assess the phenotypic genotypic characteristics of virulence antibiotic resistance genes STEC strains isolated from various types ice cream fruit juice sold Isfahan, Iran. From March 2023 2024, 500 samples-including traditional (100), industrial frozen (100)-were collected. Samples were analyzed using biochemical molecular methods for detection. Antibiotic sensitivity was evaluated disc diffusion method against 14 antibiotics. Specific primers used identify genes. Among samples, 52 (10.42%) E. positive, highest prevalence found (20%) (15%). The pathogenic subtype, particularly enterohemorrhagic (EHEC), most common (75% positive samples) (66.66%). High rates observed ampicillin (86.53%), tetracycline (76.92%), sulfamethoxazole (73.07%), while lowest recorded imipenem (7.69%). frequently detected aadA1 tetA (57.69%), sul1 (55.76%). Food contamination by pathogenic and spoilage bacteria causes approximately 47 million cases of foodborne diseases in the United States leads to tons food spoilage, worsening loss situation worldwide. Foodborne illnesses, particularly those caused by Salmonella enterica subsp. Serovar Typhimurium, present a significant challenge to public health, especially within the seafood industry due biofilm formation on foods. This study investigated antibiofilm potential of fucoidan, sulfated polysaccharide, against Typhimurium crab and shrimp surfaces. Fucoidan’s minimum inhibitory concentration (MIC) was determined be 150 µg/mL. Sub-MIC (1/8, 1/4, 1/2, MIC) were evaluated for their impact inhibition formation. Fucoidan treatment resulted in significant, dose-dependent formation, achieving 2.61 log CFU/cm2 2.45 reductions surfaces, respectively. FE-SEM analysis confirmed disruption cell membrane damage. Real-time PCR showed downregulation quorum-sensing (luxS) virulence (rpoS, avrA, hilA) genes. These results propose that fucoidan has ability as natural antibacterial agent controlling biofilms processing, thereby enhancing food safety minimizing contamination.
