ABSTRACT Background Clostridium butyricum is a probiotic widely used in animal husbandry, and there evidence to suggest that it can alleviate intestinal inflammation pigs may be related its lipoteichoic acid (LTA), but the mechanism still unclear. Objective This study aimed determine regulatory effect potential of C. LTA on LPS‐stimulated porcine epithelial line‐J2 (IPEC‐J2). Methods IPEC‐J2 cells were treated with LPS different concentrations (0.05, 0.1 0.15 mM). After treatment 0.5, 1.5 4.5 h, cell culture media collected for measurement TNF‐α IL‐10 by using ELISA kits, RT‐qPCR Western blotting detections. Further elucidating pathway regulating gene expression inhibiting key proteins toll‐like receptor antagonists C34, PDTC, SB230580 U0126. Results High‐dose significantly promoted secretion anti‐inflammatory factor cells, inhibited pro‐inflammatory short term. TLR4 reduced protein phosphorylation levels p38, ERK1/2 p65. The inhibition TLR4, p65 caused LPS; increased TLR2 expression, ERK rather than expression. Conclusion Our found was an important component inflammatory response cells. mainly while stimulating IL‐10. Vital pulp therapy is critically important for the long-term preservation of teeth with pulpitis. However, anti-inflammatory effects pulp-capping materials used in clinics remain unsatisfactory. Pulpitis tightly connected oxidative stresses; therefore, scavenging excessive reactive oxygen species (ROS) a prospective treatment. Here thermosensitive and injectable hydrogel that forms situ was developed. By using poly(d,l-lactide)-poly(ethylene glycol)-poly(d,l-lactide)-based triblock copolymer (PPP) as scaffold to carry Prussian blue nanoparticles (PBNPs), effective ROS capability via multiple enzyme-like activities, such superoxide dismutase (SOD), catalase (CAT), peroxidase (POD)activities. The developed PBNPs@PPP demonstrated anti-oxidative stress anti-inflammation capabilities vitro. As direct capping material, suppressed inflammation progression pulpitis rats while also reducing cell necrosis inflammatory infiltration tissue. RNA sequencing analyses showed functions were highly involved pathways. Through experimental verification, it found by inhibiting NF-κB/NLRP3 pathway, which reduced transmission downstream Overall, use this has potential be new, safe efficient strategy vital treat

Fibrosis is one of the major outcomes following injury in heart. Immune response niche modulates fibrosis, yet little known about how cell-autonomous immune signaling adult cardiac fibroblasts regulates fibrosis. Using FACS, single-cell sequencing from Collagen1 GFP mice and human heart failure patients, we demonstrate that TLR4 sensor expressed fibroblasts. Inhibition reduces TGF-β induced fibrotic changes such as contractibility migration treated conditions. cells show enhanced cytokine expression, inhibition expression cytokines CF, thereby reducing targets extracellular matrix genes. Thus, our data other molecules downstream are fibroblast vitro.Graphical abstract

Objective Inflammation of the surrounding environment is a major reason causing loss or injury oligodendrocyte precursor cells (OPCs) in myelin-associated diseases. Lipopolysaccharide-activated microglia can release various inflammatory factors such as tumor necrosis factor-α (TNF-α). One ways OPC death necroptosis, which be triggered by TNF-α, receptor ligand, activating receptor-interacting protein kinase 1 (RIPK1)/RIPK3/mixed lineage domain-like (MLKL) signaling pathway. This study investigated whether inhibiting ferroptosis decrease TNF-α to alleviate necroptosis. Methods Lipopolysaccharide and Fer-1 stimulate BV2 cells. The expressions GPX4 were detected western blot quantitative real-time PCR; malondialdehyde, glutathione, iron, reactive oxygen species measured assay kits. After lipopolysaccharide stimulation cells, supernatant was taken culture OPC. expression levels RIPK1, p-RIPK1, RIPK3, p-RIPK3, MLKL, p-MLKL blot. Results administration could induce decreasing marker GPX4, while inhibitor significantly increase level. prevented oxidative stress iron concentration elevation alleviated mitochondrial damage lipopolysaccharide-induced results revealed that downregulated attenuated necroptosis p-MLKL, p-RIPK3. Conclusion may potential agent for inflammation treating myelin-related

The occurrence and development of neurodegenerative diseases is related to a variety physiological pathological changes. Neuroinflammation one the major factors that induces aggravates diseases. most important manifestation neuroinflammation activation microglia. Therefore, inhibiting abnormal microglia an way alleviate neuroinflammatory In this research, inhibitory effect tabersonine (Tab) on was evaluated by establishing BV2 model induced lipopolysaccharide (LPS). It found Tab significantly inhibited production expression nitric oxide (NO), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), reactive oxygen species (ROS) in BV-2 cells stimulated LPS. addition, can also inhibit nuclear factor-κB (NF-κB) LPS, thus regulating inflammatory mediators such as inducible synthase (iNOS). These results indicated regulated release NO, IL-1β, TNF-α, IL-6 NF-κB signaling pathway, exerting its anti-neuroinflammatory effect. This first report regarding inhibition LPS-induced Tab, which drug potential for treatment
