Exosome-integrated hydrogels represent a promising frontier in bone tissue engineering, leveraging the unique biological properties of exosomes to enhance regenerative capabilities hydrogels. Exosomes, as naturally occurring extracellular vesicles, carry diverse array bioactive molecules that play critical roles intercellular communication and regeneration. When combined with hydrogels, these can be spatiotemporally delivered target sites, offering controlled sustained release therapeutic agents. ABSTRACT Skin wound repair, a highly integrated and overlapping complex process, is susceptible to infection, excessive inflammation, high oxidative stress, which can prolong the healing time or even lead chronic wound. Meanwhile, single‐layer hydrogels are unavoidably dehydrated usually due water evaporation, not conducive healing. Therefore, bilayer hydrogel dressing composed of gelatin (G) sodium alginate (A) loaded with ciprofloxacin (C) curcumin (Cur) was manufactured by extrusion 3D bioprinting technology in this study, denoted as GACCur. The experimental results showed that 50 µg/mL Cur (GACCur50) had excellent antibacterial properties antioxidative activity 54.35 ± 0.48% DPPH scavenging ratio. For highly efficient and precise drug release, transdermal delivery systems (TDDS) have recently evolved through the combination of intelligent material-based structures with various active components. Abstract Coacervates are a highly relevant class of structures formed via liquid–liquid phase separation and new coacervate‐forming polymers sought after. Here, the formation simple coacervate droplets from double hydrophilic block copolymer (DHBC) with combination poly(oligo ethylene glycol methacrylate) (POEGMA) 2‐ethyl‐2‐oxazoline (POEtOx) without use external triggers or charges is shown. At high concentration 25 wt.%, DHBC forms sizes in low micrometre range. The have relatively stability over long period time only minor coalescence observed after 4 weeks. concentrations, no coacervation observed. Background: Skin ageing, driven predominantly by oxidative stress from reactive oxygen species (ROS) induced environmental factors like ultraviolet A (UVA) radiation, accounts for approximately 80% of extrinsic skin damage. L-glutathione (GSH), a potent antioxidant, holds promise in combating UVA-induced stress. However, its instability and limited penetration through the stratum corneum hinder topical application. This study introduces novel solid lipid nanoparticle (SLN)-enriched hydrogel designed to enhance GSH stability, penetration, sustained release anti-ageing applications. Methods: GSH-loaded SLNs were prepared via double-emulsion technique optimized using factorial design. These incorporated into 1-3% (w/v) Carbopol hydrogels produce semi-solid formulation. The hydrogel's characteristics, including morphology, mechanical rheological properties, drug release, antioxidant activity, cytotoxicity, evaluated. Results: SEM FTIR confirmed uniform dispersion within hydrogel. formulation exhibited desirable gel strength (5.1 ± 0.5 g), spreadability (33.6 1.9 g·s), pseudoplasticity, elasticity. In vitro studies revealed biphasic profile, with over 72 h 70% cumulative release. significantly improved capacity, protecting human fibroblasts enhancing cell viability. Stability indicated that 4 °C was optimal storage three months. Notably, enhanced 3.7-fold. Conclusions: SLN-enriched effectively improves delivery efficacy, providing promising platform other bioactive compounds similar challenges.
