Aberrant glycosylation patterns of glycoproteins and glycolipids have long been recognized as one the major hallmarks cancer cells that has led to numerous glycoconjugate vaccine attempts. These abnormal profiles mostly originate from lack key glycosyltransferases activities, mutations, over expressions, or modifications requisite chaperone for functional folding. Due their relative structural simplicity, O -linked glycans altered mucin family particularly attractive in design tumor associated carbohydrate-based vaccines. Several such formulations generated potent monoclonal anti-carbohydrate antibodies useful diagnostic immunotherapies fight against cancer. Paradoxically, related enveloped viruses also express analogous N - patterns. However, due fact are not equipped with appropriate glycosyl enzyme machinery, they need hijack infected host cells. Although resulting very similar those normal cells, some glycan often share common simplicity identified on Consequently, given both viral glycoepitopes, vaccines could be highly generate immune responses target conditions.

Coronavirus disease 2019 (COVID-19) is caused by a new member of the Coronaviridae family known as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). There are structural and non-structural proteins (NSPs) in genome this virus. S, M, H, E proteins, NSPs include accessory replicase proteins. The NSP components SARS-CoV-2 play an important role its infectivity, some them may be pathogenesis chronic diseases, including cancer, coagulation disorders, neurodegenerative cardiovascular diseases. interact with targets such angiotensin-converting enzyme (ACE2) receptor. In addition, can stimulate pathological intracellular signaling pathways triggering transcription factor hypoxia-inducible factor-1 (HIF-1), neuropilin-1 (NRP-1), CD147, Eph receptors, which roles progression diseases like Alzheimer's disease, epilepsy, multiple sclerosis, cancers glioblastoma, lung malignancies, leukemias. Several compounds polyphenols, doxazosin, baricitinib, ruxolitinib could inhibit these interactions. It has been demonstrated that spike protein stronger affinity for human ACE2 than SARS-CoV, leading current study to hypothesize newly produced variant Omicron receptor-binding domain (RBD) binds more strongly primary strain. SARS Middle East (MERS) viruses against have become resistant previous vaccines. Therefore, review recent studies performance vaccines their effects on COVID-19 related vital need deal conditions. This examines potential initiation it anticipated serve effective vaccine or treatment Video Abstract.

Abstract In nature, lectins are widely dispersed proteins that selectively recognize and bind to carbohydrates glycoconjugates via reversible bonds at specific binding sites. Many viral diseases have been treated with due their wide range of structures, specificity for carbohydrates, ability carbohydrates. Through hemagglutination assays, these can be detected interacting various on the surface cells envelopes. This review discusses most robust rationally engineered versions, such as lectibodies, antiviral proteins. Fusion lectin antibody’s crystallizable fragment (Fc) immunoglobulin G (IgG) produces a molecule called “lectibody” act carbohydrate-targeting antibody. Lectibodies not only glycoproteins neutralize clear viruses or infected by but also perform Fc-mediated antibody effector functions. These functions include complement-dependent cytotoxicity (CDC), antibody-dependent cell-mediated (ADCC), phagocytosis (ADCP). addition entering host cells, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein S1 binds angiotensin-converting enzyme (ACE2) downregulates it type I interferons in way may lead lung disease. The SARS-CoV-2 human immunodeficiency virus (HIV) envelope heavily glycosylated, which could make them major target developing vaccines, diagnostic tests, therapeutic drugs. neutralization clearance glycans located (e.g., glycosylated protein).

Some plant foods evolve defense mechanisms to protect themselves from predators by producing inherent chemicals as secondary metabolites such cyanogenic glycosides, glycoalkaloids, glucosinolates, pyrrolizidine alkaloids, and lectins. These are beneficial for the itself but toxic other organisms, including human beings. of these believed have therapeutic benefits therefore used against chronic health complications cancer. Inversely, short- long-term exposure significant amounts phytotoxins may end up with irreversible negative problems in important organ systems, severe cases, they can be carcinogenic fatal. A systematic literature search relevant published articles indexed Google Scholar®, PubMed®, Scopus®, Springer Link®, Web Science®, MDPI®, ScienceDirect databases was obtain necessary information. Various traditional emerging food-processing techniques been found considerably reduce most toxicants food their safest level. Despite ability preserve nutritional value processed foods, processing methods limited application accessibility middle- low-income countries. As a consequence, much more work is recommended on implementation technologies, additional scientific that effective naturally occurring toxicants, particularly alkaloids.

Betacoronaviruses, responsible for the “Severe Acute Respiratory Syndrome” (SARS) and “Middle East (MERS), use spikes protruding from virion envelope to attach subsequently infect host cells. The coronavirus spike (S) proteins contain receptor binding domains (RBD), allowing specific recognition of either dipeptidyl peptidase CD23 (MERS-CoV) or angiotensin-converting enzyme ACE2 (SARS-Cov, SARS-CoV-2) cell receptors. heavily glycosylated S protein includes both complex high-mannose type N-glycans that are well exposed at surface spikes. A detailed analysis carbohydrate-binding specificity mannose-binding lectins plants, algae, fungi, bacteria, revealed that, depending on their origin, they preferentially recognize N-glycans, N-glycans. Since glycans substantially decorate proteins, mannose-specific potentially useful glycan probes targeting SARS-CoV, MERS-CoV, SARS-CoV-2 virions. Mannose-binding legume lectins, like pea lectin, monocot snowdrop lectin algal griffithsin, which specifically glycans, respectively, particularly adapted coronaviruses. biomedical prospects coronaviruses with wide-ranging including detection, immobilization, prevention, control infection.

The hallmark of acute respiratory distress syndrome (ARDS) pathobiology is unchecked inflammation-driven diffuse alveolar damage and alveolar-capillary barrier dysfunction. Currently, therapeutic interventions for ARDS remain largely limited to pulmonary-supportive strategies, there an unmet demand pharmacologic therapies targeting the underlying pathology in patients suffering from illness. complement cascade (ComC) plays integral role regulation both innate adaptive immune responses. ComC activation can prime overzealous cytokine storm tissue/organ damage. lung injury (ALI) have established relationship with early maladaptive activation. In this review, we collected evidence current studies linking ALI/ARDS dysregulation, focusing on elucidating new emerging roles extracellular (canonical) intracellular (non-canonical or complosome), (complementome) pathobiology, highlighting complementome as a vital nexus pathobiological connectome via its crosstalking other systems immunome, DAMPome, PAMPome, coagulome, metabolome, microbiome. We also discussed diagnostic/therapeutic potential future direction care ultimate goal better defining mechanistic subtypes (endotypes theratypes) through methodologies order facilitate more precise effective complement-targeted therapy treating these comorbidities. This information leads support anti-inflammatory strategy by ComC, where arsenal clinical-stage complement-specific drugs available, especially due COVID-19.
