Invasive fungal infections (IFIs) such as cryptococcal meningitis (CM) remain a serious health issue worldwide due to drug resistance closely related biofilm formation. Unfortunately, available antifungal drugs with ideal safety and promising potency are still lacking; thus, the research of new candidate therapeutic approach is urgently needed. As an important gas messenger molecule, nitric oxide (NO) shows vital inhibition on various microorganism biofilms. Hence, three series novel NO-donating azole derivatives were designed synthesized, in vitro activity well mechanism action was investigated. Among them, 3a 3e displayed excellent against Cryptococcus neoformans depending release NO. Moreover, more stable analogue 3h demonstrated markedly anti-CM effects via intranasal dropping, avoiding first-pass possessing better brain permeability bypass blood-brain barrier. These results present dropping for treatment CM, warranting further studies.

This paper discusses the mechanisms by which fungi manipulate plant physiology and suppress defense responses producing effectors that can target various host proteins. Effector-triggered immunity effector-triggered susceptibility are pivotal elements in complex molecular dialogue underlying plant-pathogen interactions. Pathogen-produced effector molecules possess ability to mimic pathogen-associated patterns or hinder binding of pattern recognition receptors. Effectors directly nucleotide-binding domain, leucine-rich repeat receptors, downstream signaling components defense. Interactions between these receptor-like kinases plants critical this process. Biotrophic adeptly exploit networks key hormones, including salicylic acid, jasmonic abscisic ethylene, establish a compatible interaction with their hosts.
