Abstract Trisomy 21 (Down syndrome, DS) is the main human genetic cause of intellectual disability (ID). Lejeune hypothesized that DS could be considered a metabolic disease, and we found subjects with have specific plasma urinary metabolomic profile. In this work confirmed alteration mitochondrial metabolism in also investigated if metabolite levels are related to cognitive aspects DS. We analyzed profiles samples from 129 46 healthy control (CTRL) by 1 H Nuclear Magnetic Resonance (NMR). Multivariate analysis NMR showed clear discrimination (up 94% accuracy) between two groups. Over the past two decades, nuclear magnetic resonance (NMR) has emerged as one of three principal analytical techniques used in metabolomics (the other being gas chromatography coupled to mass spectrometry (GC-MS) and liquid with single-stage (LC-MS)). The relative ease sample preparation, ability quantify metabolite levels, high level experimental reproducibility, inherently nondestructive nature NMR spectroscopy have made it preferred platform for long-term or large-scale clinical metabolomic studies. These advantages, however, are often outweighed by fact that most techniques, including both LC-MS GC-MS, more sensitive than NMR, lower limits detection typically 10 100 times better. Metabolomics deals with the whole ensemble of metabolites (the metabolome). NMR-based metabolomics, which emerged along with mass spectrometry techniques, is the preferred method for studying metabolites in medical research and food industries. However, NMR techniques suffer from inherently low sensitivity, regardless of their superior reproducibility. To overcome this, we made two beneficial modifications: detuned probe to reach a position called "Spin Noise Tuning Optimum" (SNTO), replaced conventional cylindrical 5 mm tube an electric field component-optimized shaped tube. We found that concerted use both modifications can increase sensitivity (signal noise ratio per unit volume) detection decrease measurement time by order magnitude. In this study, demonstrate discuss achieved signal enhancement on model non-human (bovine serum, amino acid standard mixture) human urine samples.
