Ischemic stroke (IS) is one of the principal causes disability and death worldwide. Berberine (BBR), derived from traditional Chinese herbal medicine Huang Lian, has been reported to inhibit progression stroke, but specific mechanism whereby BBR modulates ischemic remains unclear. N6-methyladenosine (m6A) modification most typical epigenetic mRNA post-transcriptional modifications, among which METTL3 common methylation transferase. During study, middle cerebral artery occlusion/reperfusion (MCAO/R) was established in mice, mice primary astrocytes neurons induced by oxygen-glucose deprivation/reoxygenation (OGD/R) simulated vitro. Level LncNEAT1, miR-377-3p detected via RT-qPCR. The levels Nampt were measured Western blot. CCK8 LDH assay performed detect cell viability. Here, we found that berberine alleviates MCAO/R-induced injury up-regulates expression astrocytes, inhibits after OGD/R, thus promoting neuronal injury. NEAT1 binds OGD/R plays a protective role as ceRNA. can enhance stability modulating m6A NEAT1. Taken together, our results demonstrate exerts neuroprotective effects methyltransferase METTL3, regulates NEAT1/miR-377-3p/Nampt axis mouse ameliorate ischemia/reperfusion (I/R)

Sirtuins are class III histone deacetylases, whose enzymatic activity is dependent on NAD+ as a cofactor. reported to modulate numerous activities by controlling gene expression, DNA repair, metabolism, oxidative stress response, mitochondrial function, and biogenesis. Deregulation of their expression and/or action may lead tissue-specific degenerative events involved in the development several human pathologies, including cancer, neurodegeneration, cardiovascular disease. The most studied member this enzymes sirtuin 1 (SIRT1), associated with increasing insulin sensitivity. SIRT1 has been implicated both tumorigenic anticancer processes, regulate essential metabolic pathways, suggesting that its activation might be beneficial against disorders metabolism. Via regulation p53 deacetylation modulation autophagy, cellular response caloric restriction lifespan extension. In recent years, scientific interest focusing identification modulators led discovery novel small molecules targeting activity. This review will examine compounds natural origin recently found upregulate activity, such polyphenolic products fruits, vegetables, plants resveratrol, fisetin, quercetin, curcumin. We also discuss potential therapeutic effects these prevention treatment disorders, particular emphasis impact.

Sirtuins are a family of proteins with enzymatic activity. There seven mammalian sirtuins (SIRT1-SIRT7) that found in different cellular compartments. They part crucial pathways and regulated by many factors, such as chemicals, environmental stress, phytochemicals. Several vitro vivo studies have presented their involvement anti-inflammatory, antioxidant, antiapoptotic processes. Recent findings imply phytochemicals resveratrol, curcumin, quercetin, fisetin, berberine, kaempferol may regulate the activity sirtuins. Resveratrol mainly activates SIRT1 indirectly AMPK. Curcumin influences SIRT3, but its is broad, cells affected. Quercetin modulates SIRT1, which triggers antioxidant responses. Fisetin, through regulation, modifies lipid metabolism anti-inflammatory Berberine has wide spectrum effects significant impact on signaling pathways. Finally, induction. This review aims to summarize recent properties modulation sirtuin activity, particular focus biochemical aspects.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) emerged as a new human pathogen in late 2019 and it has infected over 100 million people less than year. There is clear need for effective antiviral drugs to complement current preventive measures, including vaccines. In this study, we demonstrate that berberine obatoclax, two broad-spectrum compounds, are against multiple isolates of SARS-CoV-2. Berberine, plant-derived alkaloid, inhibited SARS-CoV-2 at low micromolar concentrations which was originally developed an anti-apoptotic protein antagonist, sub-micromolar concentrations. Time-of-addition studies indicated acts on the stage viral life cycle. agreement, mildly affected RNA synthesis, but strongly reduced infectious titers, leading increase particle-to-pfu ratio. contrast, obatoclax acted early infection, line with its activity neutralize acidic environment endosomes. We assessed infection primary nasal epithelial cells were cultured air-liquid interface found induced repressed expression specific sets cytokines chemokines. Moreover, both replication these target cells. propose potential could be considered further efficacy testing.

Traditionally, herbal compounds have been the focus of scientific interest for last several centuries, and continuous research into their medicinal potential is underway. Berberine (BBR) an isoquinoline alkaloid extracted from plants that possess a broad array properties, including anti-diarrheal, anti-fibrotic, antidiabetic, anti-inflammatory, anti-obesity, antihyperlipidemic, antihypertensive, antiarrhythmic, antidepressant, anxiolytic effects, frequently utilized as traditional Chinese medicine. BBR promotes metabolisms glucose lipids by activating adenosine monophosphate-activated protein kinase, stimulating glycolysis inhibiting functions mitochondria; all these ameliorate type 2 diabetes mellitus. has also shown to benefits in congestive heart failure, hypercholesterolemia, atherosclerosis, non-alcoholic fatty liver disease, Alzheimer’s polycystic ovary syndrome. investigated interesting pharmacophore with contribute significantly development novel therapeutic medicines variety disorders. Despite its enormous promise, clinical application this was severely limited because unpleasant pharmacokinetic characteristics. Poor bioavailability, absorption, poor water solubility are some obstacles restricted use. Nanotechnology suggested possible solution problems. The present review aims at recent updates on important activities different types nanocarriers used delivery diseases.

Nanomedicine has been considered a promising tool for biomedical research and clinical practice in the 21st century because of great impact nanomaterials could have on human health. The generation new smart nanomaterials, which enable time- space-controlled drug delivery, improve limitations conventional treatments, such as non-specific targeting, poor biodistribution permeability. These can respond to internal biological stimuli (pH, enzyme expression redox potential) and/or external (such temperature, ultrasound, magnetic field light) further precision therapies. To this end, photonic nanoparticles, gold, silver iron oxide, used increase sensitivity responsiveness stimuli. In review, we aim report main most recent systems that involve or stimulus-responsive release. uniqueness review lies highlighting versatility integrating these materials within different carriers. This leads enhanced performance terms vitro vivo efficacy, stability toxicity. We also point out current regulatory challenges translation from bench bedside, well yet unresolved matter regarding standardization materials.
