ABSTRACT The COVID‐19 pandemic, driven by SARS‐CoV‐2, led authorities to recommend halting assisted reproductive technology programs, focusing instead on fertility preservation, for cancer patients. presence of SARS‐CoV‐2 in semen remains controversial. This multicentric prospective cohort study, conducted across 12 university medical centers, aimed determine if is present spermatozoa/seminal plasma patients RT‐PCR and assess its impact standard parameters. levels cytokines TNF‐α were measured seminal fluid ELISA. We enrolled 129 men who underwent sperm cryopreservation between July 7, 2020, June 30, 2021. 63 included tested nasal swab samples and/or serology. All asymptomatic the day collection: 50 uninfected, 8 had a positive (PCR+) 5 seropositive. RNA was not detected or spermatozoa. Ejaculate volume significantly lower PCR+ group compared uninfected (median [IQR]: 2.6 mL [1.6–3.4] vs. 4.6 [2.6–5.2] p < 0.05). Total progressive motility seropositive (32.5% [25.0–45.0] 50% [49.0–55.0] 0.05, 22.5% [10.0; 32.5] 44.5% [40–49] Higher level observed (1.9 pg/mL [0–3.9]) (0 [0–0.4]) 0.05. Although PCR+, infection appears parameters, likely due inflammation.

Amaç: Dünya genelinde milyonlarca mortalite ve morbiditeye neden olan SARS-CoV-2 enfeksiyonu insanlarda moleküler düzeyde çok sayıda farklı patolojiye sebep olmuştur. Yapılan çalışmalar bu virüsün erkek üreme hücreleri üzerine etkili olabileceğini göstermiştir. Bu çalışmada, enfeksiyonunun miRNA’lar aracılığı ile infertiliteye nasıl olabileceğinin in silico araştırılması amaçlandı. Gereç Yöntemler: Sağlıklı testis dokusunda en yüksek oranda ifade edilen Tissue Atlas'tan elde edildi. arasından COVID-19 RNA’sını hedeflediği belirlenen miRDB veri tabanından listelendi. Elde liste makine öğrenimi algoritmasını kullanan miRWalk tabanı karşılaştırıldı. Ortak hedefler deneysel olarak doğrulanmış miRNA-hedef etkileşimleri miRTarBase sonrasında TargetScan eşleştirildi. Bulgular: RNA dizisini hedefleyen endojen seviyelerinin etkilendiği tespit dokusuna özgü belirlendi. Bunlardan hsa-miR-195-5p, hsa-miR-16-5p, hsa-miR-15a-5p, hsa-miR-15b-5p, hsa-miR-497-5p hsa-miR-424-5p’nin mir-15/16 ailesinin, hsa-miR-30c-5p, hsa-miR-30b-5p, hsa-miR-30a-5p’nin ise mir-30 ailesinin üyeleri olduğu ortaya çıkarıldı. miRNA’ların potansiyel hedef genlerinin arasında hücre döngüsü başta olmak üzere, DNA hasarı, apoptoz, spermatogenez viral cevapla ilgili ABL2, BCL2, PLEKHA1, WNK3, CCNT2, DICER1, CCND1, CCND2, CCND3 WEE1 gösterildi. Sonuç: çalışma beraber ifadesinin değişebileceğini göstermektedir, böylece erkeklerde etkisini açıklamaya yardımcı olabilir.

Animal models are indispensable for unraveling the mechanisms underlying post-acute sequelae of COVID-19 (PASC). This review evaluates recent research on PASC-related perturbations in animal models, drawing comparisons with clinical findings. Despite limited number studies post-COVID conditions, particularly those extending beyond three months, these provide valuable insights. Three hallmark features PASC-lung fibrosis, hyperglycemia, and neurological sequelae-have been successfully replicated paving way mechanistic discoveries future medical interventions. Although most have reported conditions within 14-60 days post-infection, they still offer critical reference long-term research. also explores potential persisting immune misfiring, a key factor chronicity PASC symptoms. Moreover, challenges modeling discussed, including genetic diversity inbred strains difficulties accurately identifying PASC-affected individuals. To address issues, we propose methodological improvements, such as comparing individual parameters control averages incorporating genetically diverse populations like collaborative cross models. These strategies will enhance identification characterization endotypes studies. By integrating findings from manifestations PASC, can more insights into its support development effective therapeutic strategies. Finally, emphasize urgent need longitudinal to fully uncover driving guide interventions mitigate public health impact.

A considerable proportion of males suffer from asymptomatic SARS-CoV-2 infection, while the effect on reproductive function and underlying pathomechanisms remain unclear. The total sperm count decreased evidently after yet all semen samples were tested to be RNA negative. Through label‑free quantitative proteomic profiling, a 733 proteins further identified in seminal plasma 11 COVID-19 patients seven uninfected controls. Of 37 differentially expressed proteins, 23 upregulated 14 downregulated group compared with control. Functional annotations Gene Ontology (GO), Kyoto Encyclopedia Genes Genomes (KEGG), Reactome showed that these highly enriched inflammation, immunity-related pathways as well spermatogenesis-associated biological process. Four significantly correlated one or more parameters Spearman's coefficient analysis, filtered potential hub interaction network by MCODE Cytohubba algorithms. Furthermore, we verified results Western blot analysis three representative (ITLN1, GSTM2, PSAP) validation cohort. In summary, our study acute could alter protein profile without direct testicular infection consequently lead impaired quality.
