Abstract Addressing adipose niche senescence is crucial for preventing obesity‐related aging. Telomerase reverse transcriptase (TERT) a promising target gene therapy, but traditional methods lack precision and safety. A novel mitochondrion‐located TERT (mito‐TERT) activating approach presented by injectable gene/short‐fiber complexes (gene/fiber‐plexes) to safely from mitochondrial enhancement. The gene/fiber‐plexes are prepared polydopamine‐coated short‐fibers adsorb cationic dendrimers (PAMAM G3, PG3) carrying plasmids Coenzyme Q10 (CoQ10), termed PG3‐TERT@CoQ10. Upon intraperitoneal injection, the adhere peritoneum release PG3‐TERT@CoQ10, precisely targeting niche. Transient active oxygen scavenging CoQ10 activates transfection endogenous mitochondrion translocation sequentially, enhancing function. In vitro in vivo studies shows that effectively targeted visceral tissue, increased mito‐TERT levels restored an obese mouse model, they tissue homeostasis metabolic stability. RNA sequencing indicated reduced senescence‐related genes cell cycle. This activation strategy great promise treating premature aging diseases linked senescence.

Abstract Aging is characterized by chronic low-level inflammation and associated with geriatric syndromes such as sarcopenia frailty. Our aim was to evaluate the longitudinal variation of muscle area, quality, strength, relative leukocyte-derived markers, assess presence a pathway associations among derived leukocyte ratios, components health. The InCHIANTI cohort study aging that began in 1998 follow-up visits every 3 years. Out 1 453 participants enrolled at baseline, this includes 179 complete data. Muscle strength assessed hand grip test, whereas density fat area were considered indirect markers from peripheral quantitative computed tomography calf. neutrophil-to-lymphocyte ratio (NL-ratio), age, gender, comorbidities, body mass index (BMI). over time inversely BMI, while being positively monocyte-to-lymphocyte (ML-ratio) male gender. Fat interleukine-6 (IL-6), NL-ratio, ML-ratio, BMI. Handgrip decreased IL-6 levels, but increased male, having higher In path-analysis model, ML-ratio correlates mass, density, NL-ratio only density. are may be implicated age-related mechanisms affect composition strength. These ratios represent link between immune system decline health aging. However, further studies needed identify their usefulness for early detection sarcopenia, myosteatosis, frailty older adult.

Background/Objectives: LL-37 is associated with metabolic syndrome (MetS), a constellation of risk factors comprising obesity, insulin resistance (IR), dyslipidemia, and hypertension, which elevates the cardiovascular disease type 2 diabetes. Methods: In this narrative review, we analyzed literature focusing on recent developments in relationship between cathelicidin various components MetS to provide comprehensive overview. Results: Studies have shown that linked inflammation adipose tissue (AT) development IR obesity. Cathelicidin can enhance by activating pro-inflammatory genes, as well modulate inflammatory response. The mechanisms include activation complex signaling pathways induce reduce adipocytes. Toll-like receptors (TLRs) stimulates secretion cytokines, contributing disruption function cells. also influences lipid metabolism, research showing negative levels HDL cholesterol. Therefore, involved not only regulation but potentially aggravating complications MetS. Conclusions: plays crucial role regulating balance anti-inflammatory responses Understanding impact these may unveil novel approaches for addressing its complications.

Adipose tissue exhibits a remarkable capacity to expand, contract, and remodel in response changes physiological environmental conditions. Here, we describe recent advances our understanding of how functionally distinct tissue-resident mesenchymal stromal cell subpopulations orchestrate several aspects pathophysiological adipose remodeling, with particular focus on the adaptations that occur energy surplus temperature. The study remodeling provides vehicle understand functional diversity cells offers lens through which generalizable reorganization can be readily observed.

Obesity is associated with increased DNA damage, which may in turn contribute to the development of obesity-related complications. damage can also affect adipocyte biology, resulting adiposity. Carefully managed weight loss programs reverse this process. This article surveys new data that support these contentions.

In recent decades, obesity has become a worldwide epidemic. As result, the importance of adipose tissue (AT) as metabolically active storage depot for lipids and key mediator body-wide metabolism energy balance been increasingly recognized. Emerging from studies AT in metabolic disease is recognition adipocyte progenitor cell (APC) population being gatekeeper function. APCs have capability to self-renew undergo adipogenesis propagate new adipocytes capable lipid storage, which important maintaining healthy fat pad, devoid dysfunctional droplet hypertrophy, inflammation, fibrosis, linked diseases, including type 2 diabetes. Like other dividing cells, are at risk undergoing senescence, state irreversible proliferation arrest that occurs under variety stress conditions, DNA damage telomere attrition. APC controlled by factors, paracrine endocrine quality timing intake, circadian clock system. Therefore, alteration any underlying signaling pathways resulting excessive can lead premature senescence. Better understanding senescence mechanisms will interventions extending health.

Type 2 diabetes (T2D) constitutes a major public health problem, and despite prevention efforts, this pandemic disease is one of the deadliest diseases in world. In 2022, 6.7 million patients with T2D died prematurely from vascular complications. Indeed, increases risk myocardial infarction or stroke eightfold. The identification molecular factors involved occurrence cardiovascular complications their are therefore axes. Our hypothesis that brought into play during physiological aging appear progression. study focused on extracellular matrix (ECM), element maintenance homeostasis. We characterized morphological functional aspects aorta, focus collagen elastic fibers diabetic mice aged 6 mo to nondiabetic 20 mo. comparison two models (young old) highlighted an exacerbated activity proteases, which could explain disturbance accumulation excessive degradation fibers. Moreover, generation circulating elastin-derived peptides reflects premature ECM. These elements contribute appearance rigidity, often origin pathologies such as hypertension atherosclerosis. conclusion, we show present same characteristics ECM wear those observed This accelerated aortic wall remodeling then early onset and, therefore, death T2D.

Obesity affects more than one billion people worldwide and often leads to cardiometabolic chronic comorbidities. It induces senescence-related alterations in adipose tissue, senescence is closely linked obesity. Fully elucidating the pathways through which vitamin D exerts anti-inflammatory effects may improve our understanding of local tissue inflammation pathogenesis metabolic disorders. In this narrative review, we compiled analyzed literature from diverse academic sources, focusing on recent developments provide a comprehensive overview effect associated with obesity senescence. The article reveals that activation NF-κB (nuclear factor kappa B subunit 1) NLRP3 inflammasome (nucleotide-binding domain, leucine-rich-containing, pyrin domain-containing-3) toll-like receptors, increases oxidative stress cytokine release, common mechanism underlying senescence, it discusses potential beneficial alleviating development subclinical inflammation. Investigating main target cells action could help uncover complex mechanisms cellular This review summarizes significant findings related opportunities for improving health.
