Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzheimer's disease. Ageing considered to be greatest risk factor for PD, with a complex interplay between genetics and environment. With population ageing, prevalence of PD expected escalate worldwide; thus, it utmost importance reduce burden PD. To date, there are no therapies cure disease, current treatment strategies focus on management symptoms. Older adults often have multiple chronic diseases geriatric syndromes, which further complicates Healthcare systems care models necessary address broad needs older patients largely unavailable. Parkinson's disease (PD) is the most common neurodegenerative movement disorder, and it characterized by selective loss of dopaminergic (DA) neurons in substantia nigra pars compacta (SNpc), as well presence intracellular inclusions with α-synuclein main component surviving DA neurons. Emerging evidence suggests that imbalance proteostasis a key pathogenic factor for PD. Endoplasmic reticulum (ER) stress-induced unfolded protein response (UPR) autophagy, two major pathways maintaining proteostasis, play important roles PD pathology are considered attractive therapeutic targets treatment. However, although ER stress/UPR autophagy appear to be independent cellular processes, they closely related each other. In this review, we focused on molecular cross-links between pathology. The bidirectional interaction between the gut microbiota (GM) and Central Nervous System, so-called brain axis (GMBA), deeply affects function has an important impact on development of neurodegenerative diseases. In Parkinson’s disease (PD), gastrointestinal symptoms often precede onset motor non-motor manifestations, alterations in GM composition accompany pathogenesis. The term "neurodegenerative diseases" (NDs) identifies a group of heterogeneous diseases characterized by progressive loss selectively vulnerable populations neurons, which progressively deteriorates over time, leading to neuronal dysfunction. Abstract Life expectancy has increased immensely over the past decades, bringing new challenges to health systems as advanced age increases predisposition for many diseases. One of those is burden neurologic disorders. While hypotheses have been placed explain aging mechanisms, it widely accepted that increasing pro-inflammatory status with or “inflammaging” a main determinant biological aging. Furthermore, inflammaging at cornerstone age-related diseases and its involvement in disorders an exciting hypothesis.
