Abstract BACKGROUND We sought to characterize the cognitive profile among individuals with mild impairment Lewy bodies (MCI‐LB) help guide future clinical criteria. METHODS Systematic review and meta‐analysis included MCI‐LB studies data from PubMed, Embase, Web of Science, PsycINFO (January 1990 March 2023). scores were compared controls, MCI due Alzheimer's disease (MCI‐AD), dementia (DLB) groups random‐effects models. RESULTS 26 2823 participants. Across all domains, group performed worse than controls better DLB. Compared MCI‐AD, in attention/processing speed ( g = –0.24, 95% confidence interval [CI]: –0.35, –0.12), attention/executive –0.42, CI: –0.56, –0.28); verbal immediate recall 0.37; 0.15, 0.59) delayed memory 0.40; 0.22, 0.58). DISCUSSION The profiles MCI‐AD are consistent established DLB AD. Neuropsychological assessment may be helpful differential diagnosis, even early states. Highlights a systematic for cognition (MCI‐LB). had attention, executive function, processing speed. recall. was on domains bodies. Studies used different tests there is need global efforts harmonization.

Objective The most prevalent types of dementia in older adults are Alzheimer's disease (AD) and Lewy body (LBD), they have overlapping clinical symptoms. We aimed to define amounts cortical activation identify indicators brain function better distinguish between these aid diagnosis using functional near-infrared spectroscopy (fNIRS). Abstract INTRODUCTION Males are disproportionately represented in dementia with Lewy bodies (DLB). Little is known regarding sex differences cognitive, neuropsychiatric, and motor features of the prodromal stage DLB. METHODS We used National Alzheimer's Coordinating Center longitudinal data to examine 120 males 23 females 2 years prior a clinical diagnosis RESULTS were more likely than present two or core DLB obtaining dementia. Rapid eye movement sleep behavior disorder, cognitive fluctuations, bradykinesia, rigidity common females. Neuropsychological test performance was similar between groups. DISCUSSION In DLB, exhibit greater number Core may fall short identifying at risk. Additional work needed better understand profile Highlights There presentation course neurodegenerative disease. Findings from Uniform Data Set suggest there symptoms. Females less demonstrate diagnosis. This information can help inform diagnostic criteria for

ABSTRACT OBJECTIVE Lewy body dementia (LBD) is an umbrella term describing two closely related conditions: Parkinson disease (PDD) and with bodies (DLB). LBD the second most common cause of neurodegenerative but often underrecognized in clinical practice. This review covers key epidemiologic, clinical, cognitive, behavioral, biomarker features discusses current treatment options. LATEST DEVELOPMENTS Indicative biomarkers improve ability to make a diagnosis include single-photon emission computed tomography (SPECT) dopamine system (brain) noradrenergic (cardiac), polysomnography. α-Synuclein–specific spinal fluid, skin, plasma, brain imaging are active development some available for use. Prodromal stages PDD DLB have been contextualized, diagnostic criteria published. An emerging theme whether integrated staging focusing on protein aggregation, rather than symptoms, may advance research efforts. ESSENTIAL POINTS cognitive impairment older adults subject significant delays treatment, increasing burden patients family care partners. Understanding use will recognition. Earlier detection also facilitate new therapeutics enrollment trials.
