Cannabis, the most consumed illicit psychoactive drug in world, is increasingly used by pregnant women. However, while cannabinoid receptors are expressed early embryo, impact of phytocannabinoids exposure on embryonic processes lacking. Here, we leverage a stepwise vitro differentiation system that captures developmental cascade to investigate abundant phytocannabinoid, Δ9-tetrahydrocannabinol (Δ9-THC). We demonstrate Δ9-THC increases proliferation naive mouse stem cells (ESCs) but not their primed counterpart. Surprisingly, this increased proliferation, dependent CB1 receptor binding, only associated with moderate transcriptomic changes. Instead, capitalizes ESCs’ metabolic bivalence increasing glycolytic rates and anabolic capabilities. A memory rewiring retained throughout Primordial Germ Cell-Like Cells absence direct an alteration transcriptional profile. These results represent first in-depth molecular characterization stages germline development.

SummaryCannabis consumption is legally prohibited in most countries the world. Several are legalising cannabis for adult consumption. It important to monitor public health effects of these policy changes. In this paper, we summarise evidence date on legalisation non-medical use Canada and USA. We describe regulatory models legalisation, changes products pricing, illicit market, use, cannabis-related physical mental harms. discuss challenges assessing outcomes emphasise importance continuous rigorous monitoring adverse inform design policies regulations.

Journal of Psychoactive Drugs, Год журнала: 2023, Номер 56(3), С. 342 - 352

Опубликована: Май 8, 2023

The current study examined whether sex moderates associations between emotion dysregulation (overall and six dimensions of dysregulation) problematic cannabis use. 741 adult past-month users (31.44% female) completed questionnaires on use (Marijuana Problems Scale) (Difficulties in Emotion Regulation Scale). Mann-Whitney U tests hierarchical multiple linear regressions were performed. Male reported greater difficulties with overall dysregulation, nonacceptance, goals, impulse, strategies, clarity. Overall strategies associated more severe use, relationships weaker female users. Lack emotional awareness was less male only. Examining individual differences as they relate to suggests that treatments may need be tailored for a focus specific dimensions.

Purpose: State-level prevalence data are used to investigate whether recreational cannabis legalization moderates the mediated pathway from perception of low risk harm, use, use disorder (CUD) treatment admissions, among adolescents (age 12–17) in US. Methods: Annual state measures risk, and CUD admissions between 2008 2019 (N = 542 state-year observations) were collected National Survey on Drug Use Health (NSDUH) Treatment Episode Dataset – Admissions (TEDS-A). A two-way fixed effects (state year) moderated mediation model was test indirect effect via use. Results: positive perceiving as observed prior but not afterwards. After legalization, association with strengthened, suppressed, compared before legalization. Discussion: Recreational may alter social acceptability medical self-administration cannabis, potentially leading utilization decline even factors for increase. Background Delivering prescription digital therapeutics (ie, evidence-based interventions designed to treat, manage, or prevent disorders via websites smartphone apps) in primary care could increase patient access substance use disorder (SUD) treatments. However, the optimal approach implementing remains unknown. Objective This pilot study is a precursor larger trial test whether implementation strategies (practice facilitation [PF] and health coaching [HC]) improve delivery of for SUDs care. mixed methods describes outcomes among patients 2 clinics presents qualitative findings on implementation. Methods From February 10 August 6, 2021, total 3 mental specialists embedded practices same integrated system were tasked with offering app-based SUD. In first half pilot, activities included training supportive tools. PF (at 1 clinic) HC clinics) added second half. All analyses relied secondary data, including electronic records therapeutic vendor data. Primary proportion reached by fidelity related ideal use. We used assess adherence planned barriers facilitators therapeutics. Results Of all 18 prescribed apps, (56%) downloaded app activated their prescription, 8 (44%) completed at least module content. Patients who per week average. Ideal (fidelity) was defined as completing 4 modules having monthly SUD-related visit; (6%) met these criteria weeks (of 12-week period). A 5 (28%) had prescriptions while available, (11%) successfully contacted, both declined coaching. Clinicians reported competing clinical priorities, technical challenges, logistically complex workflows part because apps required prescription. Some impacted staff turnover that coincided COVID-19 pandemic. The high engagement perception meet needs. Conclusions encountered real-world setting, especially staffing shortages, turnover, priorities clinic teams. randomized will clarify extent which Trial Registration ClinicalTrials.gov NCT04907045; https://clinicaltrials.gov/study/NCT04907045

This study investigates whether the proportion of referrals to cannabis use disorder (CUD) treatment from criminal justice system declined among adolescents (aged 12-17 years) and young adults 18-24 following state recreational (adult use) legalization in United States between 2008 2019.Cannabis referral data were extracted Treatment Episode Dataset-Admissions used calculate trends number referrals. Difference-in-differences analysis was estimate effect on state-level as a share all admissions.Nationwide, adolescent adult CUD over period. The significantly more rapidly after compared with before ( β = -0.045; 95% confidence interval, -0.079 -0.010; P 0.01). Among adolescents, trajectory decline did not change -0.033; -0.073 0.008; 0.11).The is likely due falling cannabis-related arrests. Although criminalization may result court-mandated for some without CUD, admissions during period increasing risk factors associated represents key health concern. Promoting screening other sources, such through primary care, be warranted.
