Objective In this study, we aimed to explore the potential association between COVID-19 infection, hospitalization, severe COVID-19, and erection dysfunction (ED) using two-sample Mendelian randomization (MR) method. Methods Data pertaining were extracted from latest version of Host Genetics Initiative genome-wide study (GWAS) meta-analyses (Round 7, April 2022), outcome data obtained Open GWAS database. We applied various MR analysis methods, including inverse variance weighted method, median MR-Egger regression. Results Our investigation revealed a negative causal hospitalization ED (total testosterone levels: beta = −0.026; 95% confidence interval: −0.049 −0.001). However, no evidence supported relationships for or other risk factors. Conclusion The results comprehensive suggest link total levels. Erectile Dysfunction (ED) is a prevalent sexual health condition affecting significant portion of the male population worldwide. The conventional therapeutic approaches for ED often involve use pharmaceutical agents targeting phosphodiesterase-5 (PDE5) enzyme. Currently, treatment with PDE-5 inhibitors standard approach ED, and four inhibitors, namely sildenafil, vardenafil, tadalafil, avanafil, are in use. However, these interventions may be associated adverse effects limitations. As result, there has been growing interest exploring alternative complementary options such as nutraceuticals, which bioactive compounds derived from natural sources. Nutraceuticals, include vitamins, minerals, herbs, other dietary supplements, have gained popularity their potential benefits. Certain nutraceuticals demonstrated ability to modulate various physiological pathways, including those involved erectile function.
