A radiological accident, whether from industrial, medical, or malicious origin, may result in localized exposure to high doses of ionizing radiations, leading the development local radiation injury (LRI), that evolve toward deep ulceration and necrosis skin underlying tissues. Early diagnosis is therefore crucial facilitate identification management LRI victims. Circulating microRNAs (miRNA) have been studied as potential diagnostic biomarkers several diseases including hematological defects following whole-body irradiation (WBI). This study aims identify a blood miRNA signature associated with preclinical C57BL/6J mouse model hindlimb using different 10-MV X-ray lead injuries severities. To this end, we first performed broad-spectrum plasma profiling, followed by targeted validation step, on two independent animal cohorts. Using multivariate sparse partial least square discriminant analysis, identified panel eight circulating miRNAs able segregate mice according severity. Interestingly, these were previously WBI (miR-150-5p, miR-342-3p, miR-146a-5p), inflammation (miR-18a-5p, miR-148b-3p, miR-532-5p) (miR-139-5p, miR-195-5p). Our results suggest use suitable molecular for prognosis diagnosis.

Background Different wavelengths of ultraviolet (UV) light cause skin damage through different mechanisms. Minimal erythema dose (MED) is usually used to clinically evaluate sensitivity radiation by inducing using B (UVB) or A (UVA) + UVB. UVB significantly impacts the occurrence of cutaneous disorders, ranging from inflammatory to neoplastic diseases.
