The lacrimal gland (LG) is an exocrine that produces the watery part of tear film lubricates ocular surface. Chronic inflammation, such as Sjögren’s syndrome (SS), one leading causes aqueous-deficiency dry eye (ADDE) disease worldwide. In this study we analyzed chronic inflammation in LGs NOD.B10Sn-H2b/J ( NOD.H-2b ) mice, a mouse model SS, utilizing bulk RNAseq and Visium spatial gene expression. Purpose: Loss of function the lacrimal gland (LG), which produces aqueous tear film, is implicated in age-related dry eye. To better understand this deterioration, we evaluated changes lipid metabolism and inflammation LGs from an aging model. Methods: LG sections female C57BL/6J mice different ages (young, 2–3 months; intermediate, 10–14 old, ≥24 months) were stained with Oil Red-O or Toluidine blue to detect lipids. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis western blotting lysates determined differences expression genes proteins related metabolism. A photobleaching protocol quench autofluorescence was used evaluate immunofluorescence associated NPC1, NPC2, CTSL, macrophages (F4/80, CD11b) age using confocal fluorescence microscopy. Dry eye is one of the most common ocular surface diseases caused by tear film instability and damage due to an abnormal quality or quantity tears. Inflammatory factors can initiate relevant transduction signalling pathways trigger inflammatory cascade response, resulting in inflammation. It has been shown that active ingredients Dendrobium, such as polysaccharides, alkaloids phenols, have anti-inflammatory, anti-tumour immunity-boosting effects, Dendrobium officinale extract improve glandular secretion function, increase salivary expression level water channel protein glands patients with dry syndromes. We investigated vitro cytoprotective effect extracts sodium chloride induced hyperosmotic conditions human cornea keratocytes (HKs). Results showed Kimura et Migo (DOW) loddigesii Rolfe (DLW) could upregulate aquaporins (AQP)5 protein, thus exerting a repairing promoting cell migration. Furthermore, oral administration DOW DLW enhanced production rats exerted protective on damage. AQP5 mucin (muc)5ac proteins lacrimal gland reduce response. inhibited activation corresponding mitogen-activated kinases (MAPK) NF-KB pathway, thereby playing role improving symptoms.
