Fatigue is recognized as one of the most commonly presented long-term complaints in individuals previously infected with SARS-CoV-2. This systematic review was performed to describe symptoms, etiology, possible risk factors related post-COVID-19 fatigue and therapeutic approaches used for treatment fatigue. For literature search databases PubMed, Web Science, Cochrane Library, PsycInfo were used. All articles that met inclusion criteria analyzed demographics, clinical data treatment. Included studies which focused on an adult population (18-65 years old); elderly patients chronic somatic diseases can also cause excluded. We identified 2,851, screened 2,193 finally included 20 moderate high methodological quality, encompassing 5,629 participants. Potential old age, female sex, severe status acute phase infection, a number comorbidities, prediagnosis depression/anxiety. Lastly, autoimmune etiology suspected. The coronavirus disease 2019 (COVID-19) pandemic is an ongoing global health concern, and effective antiviral reagents are urgently needed. Traditional Chinese medicine theory-driven natural drug research development (TCMT-NDRD) a feasible method to address this issue as the traditional formulae have been shown in treatment of COVID-19. Huashi Baidu decoction (Q-14) clinically approved formula for COVID-19 therapy with anti-inflammatory effects. Here, integrative pharmacological strategy was applied identify bioactive compounds from Q-14. Overall, total 343 chemical were initially characterized, 60 prototype Q-14 subsequently traced plasma using ultrahigh-performance liquid chromatography quadrupole time-of-flight mass spectrometry. Among compounds, six (magnolol, glycyrrhisoflavone, licoisoflavone A, emodin, echinatin, quercetin) identified showing dose-dependent inhibition effect on SARS-CoV-2 infection, including two inhibitors (echinatin main protease (Mpro), well (glycyrrhisoflavone A) RNA-dependent RNA polymerase (RdRp). Meanwhile, three components, licochalcone B, SARS-CoV-2-infected inflammatory cell model. In addition, glycyrrhisoflavone A also displayed strong inhibitory activities against cAMP-specific 3',5'-cyclic phosphodiesterase 4 (PDE4). Crystal structures PDE4 complex or determined at resolutions 1.54 Å 1.65 Å, respectively, both bind active site similar interactions. These findings will greatly stimulate study TCMT-NDRD

Xuanfei Baidu Formula (XBF) is an effective traditional Chinese medicine (TCM) remedy for treating coronavirus disease 2019 (COVID-19) in China. This herbal has shown effects reducing clinical symptoms and shortening the average length of hospital stay COVID-19 patients. Previous studies have demonstrated that XBF alleviates acute lung injury (ALI) by regulating macrophage-mediated immune inflammation, but mechanisms action remain elusive. study aimed to evaluate lung-protective anti-inflammatory its underlying mechanisms. Here, XBF's were investigated ALI mouse model induced inhalation atomized lipopolysaccharide (LPS). Besides, LPS-induced inflammation RAW264.7 cells was used clarify against ALI. Our results showed treatment alleviated injury, as evidenced reduced histopathological changes, pulmonary alveoli permeability, fibrosis, apoptosis tissues. In addition, decreased levels tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-1β serum bronchoalveolar lavage fluid (BALF), white blood cell (WBC) count BALF. Furthermore, consistent with vivo assay, inhibited inflammatory cytokines release pro-inflammatory polarization cells. Mechanistically, increased mitochondrial fusion upregulating Mfn1 attenuated NLRP3 inflammasome activation repressing Casp11, respectively, inhibit NF-κB MAPK pathways, thus macrophage polarization. this study, we demonstrate exerts anti-ALI -inflammatory recovering dynamics activation, providing a biological illustration efficacy

Infectious diseases have been a threat to human health globally. The relentless efforts and research enabled us overcome most of the through use antiviral antibiotic agents discovered employed. Unfortunately, microorganisms capability adapt mutate over time resistance ensues. There are many challenges in treating infections such as failure respond therapeutic agents, which has led more chronic infections, complications, preventable loss life. Thus, multidisciplinary approach collaboration is warranted create potent, effective, versatile therapies prevent eradicate old newly emerging diseases. In recent past, natural medicine proven its effectiveness against various illnesses. Most pharmaceutical currently used can trace their origin products one way, shape, or form. full potential yet be realized, numerous resources not explored analyzed. The intestine is considered to be a vital digestive organ absorb nutrients and the largest immune organ, while numerous microorganisms coexist with host. It well known that complex interactions between gut microbiota host’s system inevitably affect function of other organs, creating an “axis” them. During past few years, new technique based mainly on microfluidics cell biology has been developed emulate structure, function, microenvironment human gut, called “gut-on-chip”. This microfluidic chip provides insight into key aspects in health disease, such as gut–brain axis, gut–liver gut–kidney gut–lung axis. In this review, we first describe basic theory axis various composition parameter monitoring microarray systems, summarize development emerging advances gut–organ-on-chip, focus host-gut flora nutrient metabolism, highlight their role pathophysiological studies. addition, paper discusses challenges prospects for current further use gut–organ-on-chip platform.

The 2019 coronavirus disease (COVID-19) pandemic has become a global crisis. In the immunopathogenesis of COVID-19, SARS-CoV-2 infection induces an excessive inflammatory response in patients, causing cytokine storm severe cases. Cytokine leads to acute respiratory distress syndrome, pulmonary and other multiorgan failure, which is important cause COVID-19 progression even death. Among them, activation pathways major factor generating storms dysregulated immune responses, closely related severity viral infection. Therefore, elucidation signaling pathway providing otential therapeutic targets treatment strategies against COVID-19. Here, we discuss pathogenesis including induction, function, downstream signaling, as well existing potential interventions targeting these cytokines or pathways. We believe that comprehensive understanding regulatory dysregulation inflammation will help develop better clinical therapy effectively control diseases, such
