Background The limbus is located at a 2-mm-wide area between the bulbar conjunctiva and cornea has been suggested to be niche of corneal epithelial stem cells immune cells. Like skin intestines, also an important mucosal surface, on play critical roles in surveillance ensure barrier surface homeostasis protection from various environmental damage infections. Single-cell RNA sequencing (scRNA-seq) analysis protein tyrosine phosphatase receptor type C positive (PTPRC + ) hematopoietic could provide single cell atlas all subsets. Methods We performed single-cell generate transcriptomic profile for 804 sort-purified three healthy donors. Results Our identified primary pattern multiple subtypes, including naive T cells, antiviral effector CD8 innate such as IDO1 mature regulatory dendritic (mregDCs), macrophages, monocytes, basophils human limbus. Conclusion Overall, limbal possible contribution these adaptive response cornea.

Immunosenescence, the slow degradation of immune function over time that is a hallmark and driver aging, makes older people much more likely to be killed by common infections (such as flu) than young adults, but it also contributes greatly rates chronic inflammation in later life. Such micro nutrients are crucial for modulating effective responses their deficiencies have been associated with dysfunctional immunity elderly. In this review, we specifically focused on contribution major (Vitamins A, D E, Vitamin C; Zinc Selenium) immunomodulators ageing population especially related inflame-ageing process including autoimmunity. This review will cover these hologenomic interactions, how can modulate cell and/or cytokine production benefit hosts healthy mucous-associated along sustainable immunologic homeostasis. For example, points out modulatory effects vitamin both innate adaptive immunity, specific focus its ability suppress pro-inflammatory cytokines synthesis while enhancing regulatory T-cell function. same context, zinc described important nutrient thymic differentiation exhibits immunomodulatory functions decreasing inflammation. addition, go increase systemic low-grade and, inflammaging well actually enhance autoimmune pathologies old age. It assesses potential role additional targeted nutritional supplementation counteract effects, promoting wider resilience adults. collates current evidence highlights adequate intake autoimmunity during ageing, providing plausible origins interventions promote aging.

Splenic T cells are pivotal to the immune system, yet their function deteriorates with age. To elucidate specific aspects of cell biology affected by aging, we conducted a comprehensive multi–time point single-cell RNA sequencing study, complemented Assay for Transposase Accessible Chromatin (ATAC) and repertoire (TCR) on splenic from mice across 10 different age groups. This map age-related changes in distribution lineages functional states reveals broad composition, including prominent enrichment Gzmk+ aged mice, encompassing both CD4+ CD8+ subsets. Notably, there is marked decrease TCR diversity populations mice. We identified key pathways that may underlie perturbation functions supporting cytotoxic clonal expansion study provides insights into aging process also highlights potential targets therapeutic intervention enhance elderly. The dataset should serve as resource further research dysfunction identifying strategies.

T cells and their cell receptors (TCRs) play crucial roles in the adaptive immune system's response against pathogens tumors. However, immunosenescence, characterized by declining function quantity with age, significantly impairs antitumor immunity. Recent years have witnessed remarkable progress cell-based cancer treatments, driven a deeper understanding of biology innovative screening technologies. This review comprehensively examines maturation mechanisms, cell-mediated responses, implications thymic involution on diversity prognosis. We discuss recent advances adoptive therapies, including tumor-infiltrating lymphocyte (TIL) therapy, engineered receptor (TCR-T) chimeric antigen (CAR-T) therapy. Notably, we highlight emerging DNA-encoded library technologies mammalian for high-throughput TCR-antigen interactions, which are revolutionizing discovery novel tumor antigens optimization TCR affinity. The also explores strategies to overcome challenges solid microenvironment approaches enhance efficacy As our deepens advances, immunotherapies show increasing promise delivering durable clinical benefits broader patient population.
