Nonalcoholic fatty liver disease (NAFLD), recently redefined as metabolic-dysfunction-associated (MASLD), is liver-metabolism-associated steatohepatitis caused by nonalcoholic factors. NAFLD/MASLD currently the most prevalent in world, affecting one-fourth of global population, and its prevalence increases with age. Current treatments are limited; one important reason hindering drug development insufficient understanding onset pathogenesis NAFLD/MASLD. C-reactive protein (CRP), a marker inflammation, has been linked to NAFLD aging recent studies. As conserved acute-phase protein, CRP widely characterized for host defense functions, but link between remains unclear. Herein, we discuss available evidence involvement MASLD identify areas where further research needed. We hope this review can provide new insights into aging-associated biomarkers suggest that modulation signaling potential therapeutic target.

Background Multifaceted factors play a crucial role in the prevention and treatment of metabolic dysfunction-associated steatotic liver disease (MASLD). This study aimed to utilize multifaceted indicators construct MASLD risk prediction machine learning models explore core within these models. Methods were constructed based on seven algorithms using all variables, insulin-related demographic characteristics other indicators, respectively. Subsequently, partial dependence plot(PDP) method SHapley Additive exPlanations (SHAP) utilized explain roles important variables model filter out optimal for constructing model. Results Ranking feature importance Random Forest (RF) eXtreme Gradient Boosting (XGBoost) found that both homeostasis assessment insulin resistance (HOMA-IR) triglyceride glucose-waist circumference (TyG-WC) first second most variables. The with top 10 was superior previous PDP SHAP methods further screen best (including HOMA-IR, TyG-WC, age, aspartate aminotransferase (AST), ethnicity) model, mean area under curve value 0.960. Conclusions HOMA-IR TyG-WC are predicting risk. Ultimately, our AST, ethnicity.

With changing lifestyles, non-alcoholic fatty liver disease (NAFLD) has become the most prevalent worldwide. A substantial increase in incidence, mortality, and associated burden of NAFLD-related advanced is expected. Currently, initial diagnosis NAFLD still based on ultrasound there no approved treatment method. Lipid-lowering drugs, vitamin supplementation, lifestyle improvement treatments are commonly used clinical practice. However, lipid-lowering drugs can produce poor patient compliance specific adverse effects. Therefore, exploration bio-diagnostic markers active lead compounds for development innovative urgently needed. More more studies have reported anti-NAFLD effects mechanisms natural products (NPs), which an important source new drug to treat due their high activity low side At present, berberine silymarin been by US FDA enter phase IV studies, demonstrating potential NPs against NAFLD. Studies found that regulation lipid metabolism, insulin resistance, oxidative stress, inflammation-related pathways may play roles process. continuous updating technical means scientific theories, in-depth research targets provide possibilities find drugs. As we know, FXR agonists, PPARα dual CCR2/5 inhibitors gradually coming stage Whether exert regulating these or some unknown remains be further studied. study reviewed targets. Some works discovery docking were also discussed. It hoped this review reference values non-invasive diagnostic clinic.

Background Nonalcoholic fatty liver disease (NAFLD) stands a prevalent chronic condition significantly influenced by oxidative stress. We investigated the unclear relationship between antioxidant-rich diet and lifestyle cardiovascular (CVD) prevalence rate mortality in adult patients with NAFLD. Methods This study utilized data from National Health Nutrition Examination Survey (NHAENS) spanning 1999 to 2018 investigate association adherence an The employed Oxidative Balance Score (OBS) define antioxidant lifestyle. Results Including 8,670 NAFLD, revealed inverse OBS of most CVD conditions. Fully adjusted models demonstrated that each unit increase OBS, overall corresponded 2, 7, 2% reduction all-cause mortality, respectively. In findings Q2 Q3 were linked reduced cancer whereas did not exhibit association. Additionally, Stratified analysis age (<45 years) education level (> high school) CVD. Conclusion These results underscore protective link diminished adults particularly among younger higher-educated populations.

Metabolic dysfunction-associated steatohepatitis (MASH) is the replacement term for what used to be called nonalcoholic (NASH). It characterized by inflammation and injury of liver in presence cardiometabolic risk factors may eventually result development hepatocellular carcinoma (HCC), most common form primary cancer. Several pathogenic mechanisms are involved transition from MASH HCC, encompassing metabolic injury, inflammation, immune dysregulation fibrosis. In this context, Gas6 (Growth Arrest-Specific 6) TAM (Tyro3, Axl, MerTK) receptors play important roles. The Gas6/TAM family modulation lipid metabolism, fibrosis, tumor progression metastasis, processes which an role pathophysiology acute chronic diseases. review, we discuss MASH-associated HCC potential involvement system disease progression. addition, since therapeutic strategies limited, also speculate regarding possible future treatments involving targeting or receptors.

It is widely accepted that nine hallmarks-including mitochondrial dysfunction, epigenetic alterations, and loss of proteostasis-exist describe the cellular aging process. Adding to this, a well-described cell organelle in metabolic context, namely, lipid droplets, also accumulates with increasing age, which can be regarded as further aging-associated Independently their essential role fat stores, droplets are able control integrity by mitigating lipotoxic proteotoxic insults. As we will show this review, numerous longevity interventions (such mTOR inhibition) lead strong accumulation

Nobiletin (NOB), a flavonoid with significant antioxidant potential, holds promise for treating nonalcoholic fatty liver disease (NAFLD). In this work, we aim to assess the effects and investigate molecular mechanisms of NOB on NAFLD. After using methionine choline-deficient diet induce C57BL/6J mice, as well oleic acid HepG2 L02 cells, administered an intervention. The results indicated that significantly ameliorated lipid deposition, oxidative stress, inflammation in NAFLD both models. Its mechanism may involve Nrf2, SREBP-1c, NF-κB signaling pathways. Furthermore, Nrf2 is not only direct target improve damage but also indirectly involved lipid-lowering anti-inflammatory processes By inhibiting found regulatory role metabolism related SREBP-1c closely associated terms inflammation. Our suggest one most critical targets against multiple aspects.
