Abstract BACKGROUND Modern lifestyle has led to an increase in the age at conception. Advanced is one of critical risk factors for female-related infertility. Abstract Age-associated myometrial dysfunction can prompt complications during pregnancy and labor, which is one of the factors contributing to 7.8-fold increase in maternal mortality women over 40. Using single-cell/single-nucleus RNA sequencing spatial transcriptomics, we have constructed a cellular atlas aging myometrium from 186,120 cells across twenty perimenopausal postmenopausal women. Senescent decidual stromal cells (DSCs) are enriched in decidua during decidualization, but underlying mechanisms this process remain unclear. Here, we performed single-cell RNA transcriptomics on ESCs and DSCs found that cell senescence decidualization is accompanied by increased levels branched-chain amino acid (BCAA) transporter SLC3A2.
