Abstract Hair graying, also known as canities or achromotrichia, is a natural phenomenon associated with aging and influenced by external factors such stress, environmental toxicants, radiation exposure. Understanding the mechanisms underlying hair graying an ideal approach for developing interventions to prevent reverse age-related changes in regenerative tissues. induced ionizing (γ-rays X-rays) has emerged valuable experimental model investigate molecular pathways involved this process. In review, we examine existing evidence on radiation-induced particular focus potential role of cellular senescence. We explore current understanding aging, delve into mechanisms, highlight unique advantages using ionizing-irradiation–induced research model. By elucidating involved, aim deepen our potentially identify novel therapeutic targets address phenotypic change.

Hair graying is one of the common visible signs human aging, resulting from decreased or abolished melanogenesis due to depletion melanocyte stem cells through excess accumulation oxidative stress. Cell-free therapy using a conditioned medium (CM) mesenchymal has been highlighted in field regenerative medicine owing its potent therapeutic effects with lower regulatory hurdles and safety risk. Recently, we demonstrated that CM an immortalized cell line exfoliated deciduous teeth (SHED) protective against mouse model ulcer formation via antioxidative angiogenic activities mediated by HGF VEGF. However, date, no effective treatments for hair have developed, effect SHED-CM on remains unknown. In this study, investigated caused X-ray irradiation. Repetitive subcutaneous administrations greatly suppressed development graying, when compared control medium, reduced cutaneous expression 8-hydroxy-2′-deoxyguanosine, major product DNA damage induced reactive oxygen species. Consistent these vivo results, significantly inhibited death irradiation melanoma B16F10 cells. Immunodepletion VEGF revealed inhibition was suppression generation species, which mainly probably These results suggest activity.

This study aimed to develop a sustainable and safe alternative chemical hair dyes by exploring the functional performance safety evaluation of herbal mixtures. Natural were extracted from Lawsonia inermis leaves, Clitoria ternatea flowers, Indigofera tinctoria leaves using an eco-friendly extraction method with deionized water ultrasonication. The ratios these natural optimized statistical tools, specifically Minitab, determine most effective formulation. profiles dyeing individual their combinations evaluated focus on color intensity, stability, resistance washing light exposure. optimal mixture, ratio 2:2:1:1 (L. inermis/C. ternatea/I. tinctoria/water), demonstrated highest absorbance lowest lightness, indicating darkest profile. When applied for 30 min, this combination yielded long-lasting intense dark color. Further testing hen’s egg chorioallantoic membrane test confirmed favorable profile, examination under scanning electron microscope showed no damage cuticle, that formulation is safer than dyes. Therefore, mixture promise as synthetic dyes, further development was suggested.

This review aims to gain insight into the major causes of hair graying (canities) and how plant-derived extracts phytochemicals could alleviate this symptom. Research articles on human were searched selected using PubMed, Web Science, Google Scholar databases. We first examined intrinsic extrinsic factors associated with graying, such as reduced capacity melanin synthesis transfer, exhaustion melanocyte stem cells (MSCs) melanocytes, genetics epigenetics, race, gender, family history, aging, oxidative stress, stress hormones, systematic disorders, nutrition, smoking, alcohol consumption, lifestyle, medications, environmental factors. also various plants that have shown a potential interfere onset or progression at different levels from in vitro studies clinical studies: extract Polygonum multiflorum its components, 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside emodin; Eriodictyon angustifolium flavonoid compounds, hydroxygenkwanin, sterubin, luteolin; Adzuki beans (Vigna angularis), Fuzhuan brick tea (Camellia sinensis), Gynostemma pentaphyllum; bixin, carotenoid compound found Bixa orellana; rhynchophylline, an alkaloid certain Uncaria species. Experimental evidence supports notion plant by enhancing MSC maintenance function, reducing due physiological influences, managing secretion action hormones appropriate level. It is suggested may be reversible through following tactical approaches: selective targeting p38 mitogen-activated protein kinase (MAPK)–microphthalmia-associated transcription factor (MITF) axis, nuclear erythroid 2-related 2 (NRF2), norepinephrine–β2 adrenergic receptor (β2AR)–protein A (PKA) signaling pathway.

ABSTRACT Background Graying is an inherent and unavoidable consequence of the aging process, impacting individuals all genders. There are limited studies in Saudi Arabia that have examined prevalence predictors premature graying hair (PGH). Objectives This study aims to explore PGH before age 30 among population Arabia. Methods a cross‐sectional online survey was conducted between July 2023 February 2024 Binary logistic regression analysis used identify risk factors having gray 30. Results A total 1193 participants were involved this study. significant portion respondents reported (55.9%). The younger (younger than 44 years), smokers, those who comorbidities, anxiety, depression, family history years, dry scalp, suffer from vitamin or mineral deficiencies, loss due immune diseases (such as alopecia), use minoxidil rosemary for more likely years ( p < 0.05). Conclusion highlighted high rate associated Identified include genetic, health, lifestyle factors. Healthcare professionals decision makers advised promote awareness general public on its enhance prevention PGH. Public health initiatives campaigns smoking cessation, healthy nutrition, mental health.
